Synapses learn and remember by persistent modifications of their internal structures and composition but, due to their small size, it is difficult to observe these changes at the ultrastructural level in real time. Two-photon fluorescence microscopy (2PM) allows time-course live imaging of individual synapses but lacks ultrastructural resolution. Electron microscopy (EM) allows the ultrastructural imaging of subcellular components but cannot detect fluorescence and lacks temporal resolution. Here we describe a combination of procedures designed to achieve the correlated imaging of the same individual synapse under both 2PM and EM. This technique permits the selective stimulation and live imaging of a single dendritic spine and the subsequent localization of the same spine in EM ultrathin serial sections. Landmarks created through a photomarking method based on the 2-photon-induced precipitation of an electrodense compound are used to unequivocally localize the stimulated synapse. This technique was developed to image, for the first time, the ultrastructure of the postsynaptic density in which long-term potentiation was selectively induced just seconds or minutes before, but it can be applied for the study of any biological process that requires the precise relocalization of micron-wide structures for their correlated imaging with 2PM and EM.

Unlabelled: Forming wound canal is one of the main signs of gunshot wound. Its features are related to the following differential diagnostic signs: presence of gunshot wound, its intravitality, prescription, direction of projectile (bullet) movement, power of used weapon, etc.
Objective: To study the mechanisms of wound canal formation in gunshot injury, the pattern of damage to the biological tissues of its walls (mainly, blood vessels), the features of hemorrhages forming around it.

This study develops an integrated X-ray absorption spectroscopy (XAS) photoemission electron microscopy (PEEM) platform on beamline BL09U at the Shanghai Synchrotron Radiation Facility (SSRF), enabling nanoscale characterization of complex materials through energy-resolved imaging and local-area XAS. By using the wide range of energy tunability, full access to different polarizations and PEEM's surface sensitivity, we have established a gap-monochromator control system under the EPICS framework to synchronize the elliptically polarized undulator (EPU) gap and monochromator energy dynamically, optimizing photon flux stability for absorption fine structure analysis. Combining X-ray magnetic circular dichroism (XMCD) and X-ray magnetic linear dichroism (XMLD) with PEEM and local-area XAS, this platform achieves concurrent mapping of electronic structures and magnetic domains in ferromagnetic nano-patterns, as demonstrated through our studies of NiFe Permalloy using this system.

Rationale: Autism spectrum disorders (ASD) are a group of neurodevelopmental and multifactorial conditions with cognitive manifestations. The valproic acid (VPA) rat model is a well-validated model that successfully reproduces the behavioral and neuroanatomical alterations of ASD. Previous studies found atypical brain connectivity and metabolic patterns in VPA animals: local glucose hypermetabolism in the prefrontal cortex, with no metabolic changes in the hippocampus.

Objective: To prepare astragaloside IV dripping pills (ASDP) and assess their therapeutic effects on mice with doxorubicin hydrochloride-induced dilated cardiomyopathy (DCM).: Astragaloside IV (AS) exhibits pharmacological effects in treating cardiovascular diseases, however, its clinical application is hindered by poor solubility and low bioavailability. The study sheds light on new therapeutic strategy of DCM and development of AS formulations.
