Spirocalcaridines A and B are among the most challenging members of the marine invertebrate-derived alkaloids. Approaches to the construction and elaboration of the highly compact spirocyclic core are described. The synthesis of tricyclic guanidine via tandem oxidative amination dearomatizing spirocyclization (TOADS) using hypervalent iodine set the stage for total synthesis via the migration of the C4/C8 double bond to the C4/C5 position, followed by oxidation. The undesired but not surprising propensity of the spirocyclic cyclohexadienone to undergo rearrangement to the phenol hindered the desired olefin migration. Furthermore, initial efforts to install the oxidation sequentially, first at C5 and then at C4 in the complete carbon skeleton, were fraught with unforeseen challenges and unusual outcomes. In addition, the scope and limitations of hypervalent iodine-mediated tandem oxidative dearomatizing spirocyclization on various substrates were explored. Urethanes and thiourethanes underwent spirocyclization with an excellent yield, whereas the reaction with allylic substrates and species lacking the -methoxy substituent did not proceed. Attempts to prepare other guanidine precursors are briefly discussed.

This study aimed to analyze the amino acid composition and characterize the sequences of collagen peptides from Skipjack tuna bones (TBCPs) by liquid chromatography-tandem mass spectrometry (LC-MS/MS), and further investigate the function and mechanism of action of TBCPs in nonalcoholic fatty liver disease (NAFLD). The results showed that TBCPs contain 16 types of amino acids, among which glycine is the most abundant, and hydrophobic amino acids account for 40.75 %.

A new series of dihydronaphthalen-1(2)-one derivatives were synthesized in high yields starting from commercially available 3,5,5-trimethylcyclohex-2-en-1-one 1a, aromatic aldehydes 2, and diethyl acetylenedicarboxylate. Reaction of 1a with the aldehydes produced the respective dienones 3, which could cycloadd to dialkyl acetylenedicarboxylate, either stepwise or , under aqueous/organocatalyzed (DMAP) conditions. The respective adducts 4, were produced efficiently a Diels-Alder-double bond isomerization-oxidative aromatization sequence and were characterized based on the analysis of their H and C NMR spectra.

Background: Previous studies involving cleanup via conventional solid-phase extraction (SPE) materials to overcome matrix effects for the polar organophosphonate and -phosphinate pesticides glyphosate, glufosinate, ethephon, fosetyl, and their various metabolites often showed limitations due to the existence of various matrix compounds in plant commodities with similar polarity. To overcome existing drawbacks, we utilized the unique selectivity provided by metal oxides as SPE materials. These were exploited in a novel automated online SPE-LC-MS/MS method which allowed analyte-specific trapping in the presence of excessive amounts of matrix compounds as typically contained in extracts of the Quick Polar Pesticides (QuPPe) method.
