Proliferative retinopathy is a leading cause of irreversible blindness in humans; however, the molecular mechanisms behind the immune cell-mediated retinal angiogenesis remain poorly elucidated. Here, using single-cell RNA sequencing in an oxygen-induced retinopathy (OIR) model, we identified an enrichment of sorting nexin (SNX)-related pathways, with SNX3, a member of the SNX family that is involved in endosomal sorting and trafficking, being significantly upregulated in the myeloid cell subpopulations of OIR retinas. Immunostaining showed that SNX3 expression is markedly increased in the retinal microglia/macrophages of mice with OIR, which is mainly located within and around the neovascular tufts.

Unlabelled: Methicillin-resistant (MRSA) is a leading cause of endovascular infections, where interactions with endothelial cells play a critical role in pathogenesis. Gp05, a prophage-encoded protein, has previously been implicated in promoting antibiotic persistence by modulating MRSA cellular physiology and evading neutrophil-mediated killing. In this study, we investigated the role of Gp05 in MRSA-endothelial cell interactions, focusing on its impact on bacterial adhesion, invasion, cytotoxicity, and the host inflammatory response.

Hematopoietic stem cells (HSCs), defined as cells that can engraft an adult when transplanted, mature from precursors (pre-HSCs) that differentiate from hemogenic endothelial cells (HECs) in the embryo. Many signaling pathways required to generate the first hematopoietic stem and progenitor cells in the embryo are well-characterized, but how HSCs mature from pre-HSCs is poorly understood. Here we show that "mothers against decapentaplegic homolog 7" (SMAD7), a negative regulator of transforming growth factor beta (TGFβ) and bone morphogenetic protein (BMP) signaling, is required for pre-HSC to HSC maturation.

Carfilzomib is a second-generation proteasome inhibitor used in relapsed or refractory multiple myeloma (MM). Although effective, it can be associated with rare but life-threatening complications. We describe a 73-year-old woman with relapsed IgG-kappa MM who developed thrombotic microangiopathy (TMA) and acute kidney injury (AKI) requiring hemodialysis, three weeks after initiating a carfilzomib-based regimen.
