Cutaneous allergen sensitization (CAS) underlies atopic dermatitis (AD) and leads to various allergic symptoms, including food allergy and anaphylaxis. IL-13 expression by T follicular helper T (T) has been reported to be involved in generating high-affinity IgE antibodies and causing systemic anaphylaxis. However, the mechanisms by which IL-13 triggers IgE-mediated allergic responses remain poorly defined. In the present study, we elucidate the role of IL-13 in the CAS-mediated mechanism by which high-affinity IgE antibodies are produced when the same allergen is introduced at a distal site in the secondary sensitization. The CAS model system using mice lacking the cell lineage-specific IL-13 receptor (IL-13R) demonstrated that dendritic cells (DCs), but not T or B cells, are critical in the high-affinity IgE-mediated anaphylactic response. The IL-13 signal in type 2 conventional DCs (cDC2s) enhanced the expression of MHC class II and CD301b, which was essential for the recall of type 2 responses, inducing the production of high-affinity IgE antibodies. Similar IL-13R-expressing DCs were identified in allergic rhinitis and food allergy patients with a history of AD. These findings strongly suggest the importance of DC-specific IL-13 signaling in CAS-induced allergic reactions associated with the atopic march, which is common in human AD patients.

This research explored the effects of ginseng residue oligosaccharides (GRO-N) and ginseng polysaccharides (GP-N) on alleviating allergic rhinitis (AR). In a rat model induced by ovalbumin (OVA), both high doses of GRO-N (GRO-N-H) and GP-N (GP-N-H) significantly decreased the frequency of sneezing and rubbing behaviors in AR-affected rats. Histopathological evaluations and cytokine analyses revealed that GRO-N-H and GP-N-H notably lowered the count of goblet cells and reduced inflammatory cytokine levels in these rats.

Enhancing antibody affinity is a critical goal in antibody design, as it improves therapeutic efficacy, specificity, and safety while reducing dosage requirements. Traditional methods, such as single-point mutations or combinatorial mutagenesis, are limited by the impracticality of exhaustively exploring the vast mutational space. To address this challenge, we developed a novel computational pipeline that integrates evolutionary constraints, antibody-antigen-specific statistical potentials, molecular dynamics simulations, metadynamics, and a suite of deep learning models to identify affinity-enhancing mutations.

Immunoglobulin E (IgE) is key in the pathogenesis of allergic diseases, exerting both systemic and local effects through high-affinity binding to FcεRI on mast cells and basophils. Cross-linking of antigen-specific IgE leads to rapid degranulation and release of histamine, leukotrienes, and other mediators, resulting in classic allergic sequelae. IgE plays a key role in conditions including food allergy, atopic dermatitis, asthma, chronic spontaneous urticaria, and chronic rhinosinusitis.

Specific venom immunotherapy (VIT) in patients with hymenoptera venom allergy (HVA) represents a well-studied approach to reduce the severity of a possible anaphylactic reaction. Currently, data on mechanisms of tolerance induction at the cellular level within the first hours of therapy are lacking. To address this, total and unoccupied high-affinity IgE receptor (FcεRI) numbers per basophil, soluble FcεRI (sFcεRI) and serum tryptase levels were measured before and after the first day of VIT induction in HVA patients.

Mast cells and basophils play important roles in allergic disorders associated with specific antigens and IgE. Crosslinking of the high-affinity IgE receptor (FcεRI) by specific antigens activates several tyrosine kinases, such as Lyn and spleen-associated tyrosine kinase (Syk), resulting in the release of calcium ions (Ca) from the endoplasmic reticulum (ER) into the cytoplasm. As Ca release from the ER is essential for the release of pro-inflammatory mediators, ER stress-related molecules, such as inositol-requiring enzyme 1α (IRE1α), may play roles in mast cell and basophil activation.
