p27 is a key cell cycle gatekeeper governing the timing of Cyclin-dependent kinase (CDK) activation/inactivation and, consequently, cell proliferation. Structurally, the protein is largely unfolded, a feature that strongly increases its plasticity and interactors and enhances the number of regulated cellular processes. p27, like other intrinsically unstructured proteins, is post-translationally modified on several residues. These modifications affect its cellular localization and address p27 for specific interactions/functions. Several germline or somatic (the p27 encoding gene) mutations have been demonstrated to be associated with multiple endocrine neoplasia type 4 (MEN4), hairy cell leukemia, small-intestine neuroendocrine tumors, and breast and prostate cancers. Here, we analyzed the effect of four missense and nonsense mutations found in patients affected by MEN4 or cancers, namely, c.349C>T, p.P117S; c.397C>A, p.P133T; c.487C>T, p.Q163*; and c.511G>T, p.E171*. By transfecting breast cancer cell lines, we observed increased growth and cell motility for all the investigated mutants compared to wild-type p27 transfected cells. Furthermore, we discovered that the mutant forms exhibited altered phosphorylation on key residues and different localization or degradation mechanisms in comparison to the wild-type protein and suggested a possible region as crucial for the lysosome-dependent degradation of the protein. Finally, the loss of p27 ability in blocking cell proliferation was in part explained through the different binding efficiency that mutant p27 forms exhibited with Cyclin/Cyclin-dependent Kinase complexes (or proteins involved indirectly in that binding) with respect to the WT.

Injections have been linked to feline sarcomas (feline injection-site sarcoma; FISS) and cutaneous lymphomas (cutaneous lymphoma at injection site; CLIS). Both tumors often exhibit lymphoplasmacytic inflammation ascribed to injected immunogenic material. CLIS is hypothesized to emerge from transformation and clonal expansion of lymphoid cells following persistent immune stimulation with feline leukemia virus (FeLV) reactivation and transformation.

Hydrogel-encapsulation allows slow and stable delivery of drugs with short half-lives, avoiding the undesired side effects of high-dose or frequent administration of drugs. We previously reported that gavage or injection of caffeic acid phenethyl ester (CAPE) at 10-15 mg/kg, 2-3 times per week effectively repressed the tumor growth of human prostate xenografts in nude mice. However, daily oral or injectable delivery of CAPE in prostate cancer (PCa) patients may be impractical due to forgetfulness, physical limitations, or treatment fatigue.

Purpose: Pretreatment specimens from patients treated on the I-SPY2 neoadjuvant breast cancer trial were studied to identify prespecified biomarkers associated with response to the regimen of paclitaxel, the anti-type I insulin-like growth factor receptor (IGF-1R) antibody ganitumab, and metformin (PGM) followed by doxorubicin and cyclophosphamide (AC) compared with control therapy (paclitaxel followed by AC). The primary endpoint of this trial is pathologic complete response (pCR).
Experimental Design: One hundred six patients treated with PGM and 119 contemporary controls were evaluated using laser capture microdissection and reverse-phase protein array to evaluate 32 prespecified potential predictive biomarkers in the IGF-1R pathway and 109 additional exploratory endpoints.

ObjectivesHistorically, vaccines have been administered in the dorsal interscapular region of cats (the 'scruff' of the neck) owing to easy access to the subcutaneous space. In response to concerns about sarcomas developing at injection sites (feline injection site sarcomas [FISSs]), and a possible association between feline leukaemia virus (FeLV) vaccination and the development of FISS, alternative FeLV vaccination sites such as the distal left hindlimb and tail have been proposed by influential vaccination bodies and various key opinion leaders. There is a dearth of evidence, however, to demonstrate the development of a comparable immune response after FeLV vaccination in these sites.
