Adenovirus (Adv) is increasingly recognized for its significance in the fields of gene therapy and viral vector vaccines. The diverse applications in clinical trials and fundamental research necessitate the development of environmentally and economically sustainable purification processes that are straightforward and scalable for both academic and industrial contexts. In the initial segment of this study, we evaluated the lysis efficiency of polysorbate 20 (PS20) in comparison to polysorbate 80 (PS80). Our findings indicated that the viability HEK293 could be reduced to approximately 13 %, with a detectable Adv5 concentration of average 1.62 × 10 ifu/mL in the supernatant after an incubation in 1.0 % (w/w) PS20 or PS80 buffer. In the subsequent portion of this research, we employed a high-throughput static binding capacity (SBC) screening tool in conjunction with on-column dynamic binding capacity (DBC) validation to concurrently assess the binding capacity of Adv5 on nine different types of anion exchange media. The results demonstrated that both Sartobind Q membrane and POROS 50HQ resin exhibited binding capacities exceeding 1.0 × 10 vp/mL under the testing conditions.

Pemphigus vulgaris (PV) is an autoimmune blistering disorder, which is caused by the loss of desmosomal cell-cell adhesion, initiated by the binding of IgG antibodies against the desmosomal components desmoglein (Dsg)1 and Dsg3. Dsg3-reactive CD4 T helper (Th) cells, in particular follicular Th (Tfh) cells, play a central role in autoantibody production by Dsg3-specific B cells. In this study, we challenged the concept that distinct Dsg3-reactive CD4 T cell subsets are critical in PV pathogenesis utilizing phenotypical and functional state-of-the-art ex vivo assays.

Parvalbumin (PV), a thermostable and digestion-resistant fish allergen, has been shown to retain its allergenic potential following traditional treatments, thus posing a persistent allergic risk. The study investigated the digestive kinetics and IgE immunoreactivity of Trachinotus ovatus PV, a major fish allergen, under different treatments (untreated; DPCD treatment-15 MPa, 30 min, 50 °C; heat treatment), to evaluate its allergenic potential alterations. The analysis was conducted using a combination of techniques to assess the proteolytic stability and IgE-binding capacity of PV, including Tris-Tricine-SDS-PAGE, Western blot (WB), indirect enzyme-linked immunosorbent assay (ELISA), and free amino group quantification.

Phage display libraries of human single-chain variable fragments (scFv) serve as a valuable resource for generating fully human antibodies for scientific and clinical applications. In this study, we designed and constructed a highly diverse semi-synthetic humanized scFv phage display library using an optimized Kunkel mutagenesis approach. Our optimizations eliminated residual template, enhancing mutagenesis efficiency and expanding library diversity with a reservoir capacity exceeding 10.
