Aldolases, especially 2-deoxyribose-5-phosphate aldolase (DERA) enzymes, have been widely employed to access key chiral precursors for various active pharmaceutical ingredients (APIs). This has been enabled by expanding their substrate scope toward non-natural acceptors and donors via protein engineering. In this study, we endeavored to broaden the acceptor substrate scope of DERA from sp. (DERA) toward the heteroaryl aldehydes through a rational protein engineering approach. We successfully performed iterative saturation mutagenesis of DERA, resulting in two enantiocomplementary variants, viz., ()-selective DERA-S185G and ()-selective DERA-T12I/S185A, with enhanced catalytic efficiencies and enantioselectivities. Remarkably, the natural enantioselectivity of DERA was reversed by a single mutation (S185G). The synthetic applicability of the mutants was demonstrated by conducting aldol reactions on a semipreparative scale, from which both ()- and ()-enantiomers of heteroaryl aldols were isolated with high yields (up to 99%) and excellent enantiopurities (up to 99:1 e.r.).

A direct azidomethylation reaction at the sulfur atoms of thiols with -azidomethyldisulfonimides is presented, providing a facile and efficient approach for the synthesis of azidomethylated compounds with broad substrate scope and mild reaction conditions. Under optimized conditions using -azidomethyl-bis(4-trifluoromethylbenzene)sulfonimide as the azidomethyl source, various aliphatic and aromatic thiols furnish the corresponding -azidomethyl compounds in moderate to high yields. The reaction proceeds selectively at the mercapto group, even in substrates bearing polar functional groups.

The carbonyl-ene reaction between aldehydes and olefins constitutes a perfectly atom economic approach to homoallylic alcohols with concomitant C-C bond formation. However, the scope of catalytic asymmetric intermolecular versions is currently limited to activated substrates, while of the two intramolecular types only catalytic asymmetric (-)-carbonyl-ene type cyclizations can be considered mature. The corresponding (-)-cyclizations would arguably find equal utility in chemical synthesis but remain underdeveloped.

Herein, we have developed a Brønsted acid catalyzed 1,5-migration of functional groups from indole-tethered ynamides to prepare a variety of 2-acyltryptamines in good to excellent yields with high site-selectivity at the C2-position of indoles. Mechanistic studies revealed that the reaction underwent an intramolecular cyclization, 1,2-migration of the vinyl group, and C-N bond cleavage by hydrolysis in a one pot. The reaction features broad substrate scope, good functional group compatibility, 1,5-migration of functional groups, C-N bond cleavage to form C-C bond, and diverse 2-acyltryptamine scaffolds.

A regioselective C2-alkynylation of indoles ruthenium(II)-catalyzed C-H activation using bromoalkynes is demonstrated under both solution-phase and mechanochemical conditions. The solvent-minimized mechanochemical method delivers comparable yields with reduced reaction time and improved green metrics. Broad substrate scope, gram-scale applicability, and post-functionalization showcase the synthetic utility of this approach.

Under visible-light photocatalysis facilitated by cobalt coordination, a highly regio- and stereoselective cycloisomerization reaction of 1,6-enynes has been developed. This method enables the efficient synthesis of various skipped 1,4-diene products with excellent stereoselectivity, using commercially available cobalt catalysts, ligands, and reagents. Notably, the reaction exhibits remarkable regioselectivity (>20:1), stereoselectivity (/ > 20:1), and high yields (58-92%) under mild conditions, along with a broad substrate scope and good functional group tolerance.
