Article Abstract

Cryo-EM particle identification from micrographs ("picking") is challenging due to the low signal-to-noise ratio and lack of ground truth for particle locations. State-of-the-art computational algorithms ("pickers") identify different particle sets, complicating the selection of the best-suited picker for a protein of interest. Here, we present REliable PIcking by Consensus (REPIC), a computational approach to identifying particles common to the output of multiple pickers. We frame consensus particle picking as a graph problem, which REPIC solves using integer linear programming. REPIC picks high-quality particles even when the best picker is not known a priori or a protein is difficult-to-pick (e.g., NOMPC ion channel). Reconstructions using consensus particles without particle filtering achieve resolutions comparable to those from particles picked by experts. Our results show that REPIC requires minimal (often no) manual intervention, and considerably reduces the burden on cryo-EM users for picker selection and particle picking. Research Question: Can embryologists reliably differentiate euploid from aneuploid embryos based on morphology alone, and can artificial intelligence (AI)-assisted selection, specifically using ERICA (Embryo Ranking Intelligent Classification Algorithm), improve embryo selection outcomes compared with embryologists alone?

Design: A training tool was developed and used in which 19 embryologists (comprising junior, intermediate and experienced practitioners) evaluated the ploidy status of embryo images. They were subsequently provided with rankings generated by ERICA for the same embryos and asked to make a final judgment combining both sources of information. Each embryologist conducted this process on between 20 to 150 simulated IVF cycles to assess performance in identifying euploid embryos.

To achieve accurate detection of the pineapple fruit picking area and pose under complex backgrounds and varying lighting conditions, this study proposes a pineapple keypoint detection model (LTHRNet) based on an improved LiteHRNet. Image data of pineapple fruits under different lighting conditions were collected, and six keypoints were defined to characterize the morphological features of the fruit. In the model design, LTHRNet incorporates the LKA_Stem module to enhance initial feature extraction, the D-Mixer module to capture both global and local feature relationships, and the MS-FFN module to achieve multi-scale feature fusion.
