Protein folding is a complex process influenced by the primary sequence of amino acids. Early studies focused on understanding whether the specificity or the conservation of properties of amino acids was crucial for folding into secondary structures such as α-helices, β-sheets, turns, and coils. However, with the advent of artificial intelligence (AI) and machine learning (ML), the emphasis has shifted towards the precise nature and occurrence of specific amino acids. In our study, we analyzed a large set of proteins from diverse organisms to identify unique features of secondary structures, particularly in terms of the distribution of polar, non-polar, and charged amino acid residues. We found that α-helices tend to have a higher proportion of charged and non-polar groups compared to other secondary structures and that the presence of oppositely charged amino acid residues in helices stabilizes them, facilitating the formation of longer helices. These characteristics are distinct to α-helices. This study offers valuable insights for researchers in the field of protein design, enabling the de-novo creation of short helical peptides for a range of applications. We have also developed a web server for extensive analysis of proteins from different databases. The web server is housed at https://proseqanalyser.iitgn.ac.in/.

B cells are critical components of the adaptive immune system that proliferate and differentiate within the secondary lymphoid organs upon recognition of antigens and engagement of T cells. Traditional two-dimensional (2D) cell cultures fall short of replicating the intricate structures and dynamic evolution of three-dimensional (3D) environments found in lymphoid organs, prompting the development of more physiologically pertinent models. Our approach employs -hexanoyl glycol chitosan (HGC) coated ultra-low attachment (ULA) lattice plates to cultivate a 3D co-culture of CD40L-expressing MS5 stromal cells and naïve B cells derived from the peripheral blood mononuclear cells (PBMCs) of healthy human donors.

Investigation of the fundamental microscopic processes occurring in organic reactions is essential for optimising both organocatalysts and synthetic strategies. In this study, single-molecule fluorescence microscopy was employed to study the Diels-Alder reaction catalysed by a first-generation MacMillan catalyst, providing direct insights into its kinetic dynamics. The analytical comparability of biologic products and their biosimilars, including higher-order structure (HOS) assessment, ensures product quality and is required for regulatory approval. In this study, nuclear magnetic resonance (NMR) spectroscopy was used to evaluate the HOS of Humira (adalimumab) and its biosimilars under normal and photo-stressed conditions. Under normal conditions, 1D and 2D NMR spectra showed strong structural similarity among all products.

Relations between children's motor skills and internalizing problems are poorly understood. The environmental stress hypothesis (ESH), originally developed for motor-impaired children, may provide understanding, yet has been scarcely examined in typically developing children. Therefore, we examined: (1) relations between children's motor skills and internalizing problems; (2) the role of secondary stressors, specifically interpersonal conflicts and externalizing problems; and (3) the role of personal resources, namely, prosocial behaviour and social self-efficacy (SSE).
