Glioblastoma multiforme (GBM) is an aggressive brain cancer with a high mortality rate and limited treatment options. Metabolism-based synergistic therapy holds promise for GBM treatment, however, its efficacy is significantly impeded by poor blood-brain barrier (BBB) penetration, inadequate targeting of GBM cells, and systemic drug-related side effects. To address these challenges, we herein developed a dual-targeting nanoplatform, EVs@siMCT4-HFn@AuMn, by arming siMCT4-loaded M1-type microglia-derived extracellular vesicles (EVs) with ultrasmall nano-Au/MnO-loaded H-ferritin (HFn).

As a major fuel source for the tricarboxylic acid cycle, lactate controls energy metabolism through cell-to-cell or tissue-to-tissue lactate shuttles via monocarboxylate transporters (MCTs). Although lactate is shown to influence macrophage functions via histone lactylation, the specific functions of MCTs in macrophages remain incompletely understood. This study discovers that MCT1 and MCT4 have contrasting effects on regulating macrophage polarization.

Hypoxia-inducible factor (HIF)-1α is a master transcription factor regulating hypoxic adaptation and activates the transcription of genes involved in various steps of energy metabolism. However, some subsets of cancer cells exhibit high HIF-1α levels regardless of the oxygen concentration. Even under normoxic and normoglycemic conditions, HIF-1α regulates basal expression of its target genes.

Head and neck squamous cell carcinoma (HNSCC) exhibits a poor prognosis, particularly in advanced stages characterized by high recurrence and metastasis rates. This study investigates the role of lactate metabolism in HNSCC, aiming to develop a prognostic model to predict immunotherapy outcomes. Genomic and clinical data from The Cancer Genome Atlas and Gene Expression Omnibus databases were analyzed, focusing on 233 lactate metabolism-related genes (LMGs).

Introduction: (MAP) plays a significant role in Crohn's disease (CD). Monocarboxylate transporter 4 (MCT4) is a proton-coupled symporter of lactate that facilitates the inflammatory shift in macrophages and increases their reliance on glycolysis. MCT4 is also involved in the negative regulation of intestinal epithelial barrier function.
