Correlative light and electron microscopy (CLEM) pipelines serve to integrate the imaging modalities of fluorescence light microscopy (FLM) and cryogenic electron microscopy (cryo-EM) to produce contextually relevant high-resolution structural snapshots of biological systems. Innovations in sample preparation, instrumentation, imaging, and data processing have advanced the field of cryo-EM. This review focuses on prior work and recent developments in the field of cryo- EM that support further integration of technologies for correlative microscopy workflows.

Background: Amyotrophic lateral sclerosis (ALS) is a devastating neurodegenerative disease characterized by oxidative stress and progressive motor neuron degeneration. This study evaluates the potential neuroprotective effects of caffeine in the Wobbler mouse, an established model of ALS.
Methods: Wobbler mice received caffeine supplementation (60 mg/kg/day) via drinking water, and key parameters, including muscle strength, NAD metabolism, oxidative stress, and motor neuron morphology, were assessed at critical disease stages.

Insect chemoreception is essential for locating food, selecting oviposition sites, and detecting infochemicals. In tephritid fruit flies, chemosensory perception occurs primarily through sensilla on the antennal flagella, maxillary palps, and ovipositor. Identifying these sensilla provides insights into olfaction, which may lead to improvements in insect control measures.

A CoO/AgMoO/CeOternary nanocomposites photocatalyst was successfully synthesized through a straightforward ethanol-assisted chemical method. Comprehensive characterization of its structural and optical properties was conducted using X-ray diffraction (XRD), Fourier-transform infrared spectroscopy (FTIR), Raman spectroscopy, scanning electron microscopy (SEM), transmission electron microscopy (TEM), UV-Vis diffuse reflectance spectroscopy (UV-DRS), and photoluminescence (PL) analysis. XRD analysis confirmed the presence of CoO, AgMoO and CeO in the ternary composite sample.

Volume correlative light and electron microscopy (vCLEM) is a powerful imaging technique that enables the visualization of fluorescently labeled proteins within their ultrastructural context. Currently, vCLEM alignment relies on time-consuming and subjective manual methods. This paper presents CLEM-Reg, an algorithm that automates the three-dimensional alignment of vCLEM datasets by leveraging probabilistic point cloud registration techniques.

As a key mitochondrial Ca transporter, NCLX regulates intracellular Ca signalling and vital mitochondrial processes. The importance of NCLX in cardiac and nervous-system physiology is reflected by acute heart failure and neurodegenerative disorders caused by its malfunction. Despite substantial advances in the field, the transport mechanisms of NCLX remain unclear.
