Article Abstract

Ingestible microdevices represent a breakthrough in non-invasive sampling of the human gastrointestinal (GI) tract. By capturing the native spatiotemporal microbiome and intricate biochemical gradients, these devices allow a non-invasive multi-omic access to the unperturbed host-microbiota crosstalk, immune/nutritional landscapes and gut-organ connections. We present the current progress of GI sampling microdevices towards personalized metabolism and fostering collaboration among clinicians, engineers, and data scientists.

Herein, we introduce a LEGO®-inspired molecular diagnostic microdevice fully integrated with DNA extraction, loop-mediated isothermal amplification (LAMP), and colorimetric detection functionalities for rapid detection of antibacterial resistance in a pipette-free manner. The microdevice system is composed of a ready-to-use microdevice containing all necessary reagents and stamps that offer sample-to-answer diagnosis in a pipette-free manner. In particular, antimicrobial resistance was analyzed through LAMP at a significantly reduced temperature of approximately 40 °C, combined with DNA extraction and detection, which were performed at room temperature (RT).

Background: We recently described the utility of the "TB Concentration & Transport" kit for biosafe, ambient temperature transport of dried sputum on -Filter, and the "TB DNA Extraction" kit for DNA extraction from -Filter for the early diagnosis of drug-resistant tuberculosis (TB). This study aimed to assess the feasibility and compatibility of these kits with line probe assay (LPA) under National Tuberculosis Elimination Programme (NTEP) settings.

Methods: Patients with presumptive pulmonary TB, multidrug-resistant (MDR) TB, or extensively drug-resistant TB (N = 8491) who attended Designated Microscopy Centers (DMCs, n = 13) under National Reference Laboratories (NRLs) at Bhopal, New Delhi, Chennai, and Bhubaneswar were screened by smear microscopy.
