Article Abstract

Microbubble-enhanced ultrasound provides a noninvasive physical method to locally overcome major obstacles to the accumulation of blood-borne therapeutics in the brain, posed by the blood-brain barrier (BBB). However, due to the highly nonlinear and coupled behavior of microbubble dynamics in brain vessels, the impact of microbubble resonant effects on BBB signaling and function remains undefined. Here, combined theoretical and prospective experimental investigations reveal that microbubble resonant effects in brain capillaries can control the enrichment of inflammatory pathways that are sensitive to wall shear stress and promote differential expression of a range of transcripts in the BBB, supporting the notion that microbubble dynamics exerted mechanical stress can be used to establish molecular, in addition to spatial, therapeutic windows to target brain diseases. Consistent with these findings, a robust increase in cytotoxic T-cell accumulation in brain tumors was observed, demonstrating the functional relevance and potential clinical significance of the observed immuno-mechano-biological responses.

This paper examines how water temperature affects the dynamics of a single cavitation bubble in free field conditions. Both experimental and theoretical approaches are employed to explore the bubble dynamics in water under different temperatures. A series of single bubble experiments are conducted in water using the capacitive discharge method, with water temperature ranging from room temperature to near boiling point under atmospheric pressure.

In this review, the theoretical studies are presented for the microcavitation bubble dynamics problems, which mainly depend on the microcavitation models, such as the Rayleigh-Plesset model, the Church model, the diffusion-concentration model, and the Keller-Miksis model in biological systems. The various solutions to these models, which were formulated based on basic mathematical and physical concepts, are schematically presented. Moreover, these models are employed in many different physical problems, such as the treatment of cancerous tumours via a technique known as histotripsy and lipid shells of membrane cells, that employ focused ultrasonic therapy as a non-invasive tissue ablation method.

Ultrasound-guided drug and gene delivery (usdg) enables controlled and spatially precise delivery of drugs and macromolecules, encapsulated in microbubbles (embs) and nanoscale gas vesicles (gvs), to target areas such as cancer tumors. It is a noninvasive, high precision, low toxicity process with drastically reduced drug dosage. Rheological and acoustic properties of gvs and embs critically affect the outcome of usdg and imaging.

Unlabelled: : To assess the role of brain hemodynamics in neurodegenerative diseases, current imaging technologies remain insufficient due to limitations in spatial or temporal resolution for quantitative mapping of pulsatile flow in the whole brain. This study aims to demonstrate the feasibility of 3D transcranial Dynamic Ultrasound Localization Microscopy (DULM) for spatiotemporal blood flow measurements in the brain, addressing limitations of 2D imaging for velocity estimation within the 3D complex vascularized structures.
