Article Abstract

Telomeric G-quadruplexes (G4s) are non-canonical DNA structures composed of TTAGGG repeats. They are extensively studied both as biomolecules key for genome stability and as promising building blocks and functional elements in synthetic biology and nanotechnology. This is why it is extremely important to understand how the interaction between G4s is affected by their topology. We used small-angle x-ray scattering to investigate the end-to-end stacking of antiparallel telomeric G-quadruplexes formed by the sequence AG3(T2AG3)3. To represent the experimental data, we developed a highly efficient coarse-grained fitting tool, which successfully described the samples as an equilibrium mixture of monomeric and dimeric G4 species. Our findings indicate that the antiparallel topology prevents the formation of long multimeric structures under self-crowding conditions, unlike the hybrid/parallel structures formed by the same DNA sequence. This result supports the idea that the stacking of monomeric G-quadruplexes is strongly affected by the presence of diagonal loops.

Kaempferol (3,4',5,7-tetrahydroxyflavone, KMP) and curcumin (diferuloylmethane, CUR) are naturally occurring polyphenolic compounds with broad therapeutic potential, including anticancer, antioxidant, and anti-inflammatory properties. Among their molecular targets, DNA plays a central role, particularly through interactions with non-canonical DNA structures such as G-quadruplexes (G) and i-motifs (C), which form in guanine- and cytosine-rich genomic regions, respectively. These structures regulate telomere maintenance, gene expression, and genomic stability, making them attractive drug targets.

Telomeric G-quadruplexes (G4s) represent intriguing targets for tumours characterized by the Alternative Lengthening of Telomere (ALT) mechanism. Here we have investigated the effects of two naphthalene diimide (NDI)-based G4 interacting agents (NMe2 and QN-302) in a pair of ALT-positive human osteosarcoma (U-2 OS and Saos-2) cell lines. Both NDIs displayed marked cell growth inhibitory activity associated with the induction of telomere dysfunctions.

cGAS-STING signaling is a critical mechanism responsible for detecting cytosolic DNA, among a variety of pathways activating innate immunity, making it as an important target for cancer immunotherapy. G-quadruplexes (G4s) frequently forming in telomeres and promoters are able to regulate biological processes. Many compounds targeting G4s are reported with the capability of triggering DNA damage, while leakage of damaged DNA to cytoplasm can be the signal to stimulate innate immunity.
