Immune checkpoint inhibitors have revolutionized cancer therapy and are increasingly used to treat a wide range of oncological conditions, with dramatic benefits for many patients. Unfortunately, the resulting increase in T cell effector function often results in immune-related adverse events (irAEs), which can involve any organ system, including the central nervous system (CNS) and peripheral nervous system (PNS). Neurological irAEs involve the PNS in two-thirds of affected patients. Muscle involvement (immune-related myopathy) is the most common PNS irAE and can be associated with neuromuscular junction involvement. Immune-related peripheral neuropathy most commonly takes the form of polyradiculoneuropathy or cranial neuropathies. Immune-related myopathy (with or without neuromuscular junction involvement) often occurs along with immune-related myocarditis, and this overlap syndrome is associated with substantially increased mortality. This Review focuses on PNS adverse events associated with immune checkpoint inhibition. Underlying pathophysiological mechanisms are discussed, including antigen homology between self and tumour, epitope spreading and activation of pre-existing autoreactive T cells. An overview of current approaches to clinical management is provided, including cytokine-directed therapies that aim to decouple anticancer immunity from autoimmunity and emerging treatments for patients with severe (life-threatening) presentations.

This study aimed to identify brain activity modulations associated with different types of visual tracking using advanced functional magnetic resonance imaging techniques developed by the Human Connectome Project (HCP) consortium. Magnetic resonance imaging data were collected from 27 healthy volunteers using a 3-T scanner. During a single run, participants either fixated on a stationary visual target (fixation block) or tracked a smoothly moving or jumping target (smooth or saccadic tracking blocks), alternating across blocks.

Alpha oscillations have been implicated in the maintenance of working memory representations. Notably, when memorised content is spatially lateralised, the power of posterior alpha activity exhibits corresponding lateralisation during the retention interval, consistent with the retinotopic organisation of the visual cortex. Beyond power, alpha frequency has also been linked to memory performan ce, with faster alpha rhythms associated with enhanced retention.

Over three decades, statistical parametric mapping has transformed neuroimaging from descriptive mapping to causal inference, placing generative models at the core of causal explanations for brain function. It inspired to a large degree The Virtual Brain, which builds subject-specific digital twins from multimodal data, enabling brain simulations and exploration. Both frameworks converge at parameter estimation, where model and data meet, providing the mathematical manifestation of cause-effect in pathophysiology.

From the early development of neurometrics in Cuba to global initiatives like the Global Brain Consortium, our trajectory has paralleled and intersected with that of SPM. We highlight shared commitments to generative modeling, Bayesian inference, and population-level brain mapping, as shaped through collaborations, workshops, and joint theoretical work with Karl Friston and his group.
