Article Abstract

Photocatalysis holds a pivotal position in modern organic synthesis, capable of inducing novel reactivities under mild and environmentally friendly reaction conditions. However, the merger of photocatalysis and transition-metal-catalyzed asymmetric C-H activation as an efficient and sustainable method for the construction of chiral molecules remains elusive and challenging. Herein, we develop a cobalt-catalyzed enantioselective C-H activation reaction enabled by visible-light photoredox catalysis, providing a synergistic catalytic strategy for the asymmetric dearomatization of indoles with high levels of enantioselectivity (96 % to >99 % ee). Mechanistic studies indicate that the excited photocatalyst was quenched by divalent cobalt species in the presence of Salox ligand, leading to the formation of catalytically active chiral Co(III) complex. Moreover, stoichiometric reactions of cobaltacycle intermediate with indole suggest that the irradiation of visible light also play a critical role in the dearomatization step.

The enantioselective [4 + 4] cycloaddition for the construction of cyclooctanoids is a challenging transformation in organic chemistry. Herein, we present the first organocatalytic enantioselective [4 + 4] cycloaddition of furan ortho-quinodimethanes, activated by dearomatization of the heteroaromatic compound, which thereby allows for the cycloaddition with dienes. The [4 + 4] cycloaddition is catalyzed by a quinine-derived primary amine in combination with a chiral phosphoric acid and a carboxylic acid affording cyclooctanoids isolated as a single diastereoisomer in good yields and with up to 94% ee.

Cyclobutane-fused heterocycles are important motifs in biologically active molecules, yet their enantioselective synthesis remains a significant challenge. We report a broadly applicable and modular strategy for constructing these strained architectures through a visible-light-mediated, Lewis acid-catalyzed dearomative [2+2] photocycloaddition of indoles, benzofurans, and benzothiophenes with alkenes. The method employs a simple catalytic system based on commercially available rare-earth Lewis acids and chiral pyridine-2,6-bis(oxazoline) (PyBox) ligands.

Harnessing Noncovalent π‑Type Interactions in Thiourea-Chloride Supramolecular Complexes: Toward the Asymmetric Dearomatization of Diazaheterocycles.

Enantioselective dearomatization of diazaheterocycles through anion-binding catalysis has been developed. The process involves the nucleophilic addition of phosphorus nucleophiles to generated -benzoyliminium chlorides, using a -leucine-derived thiourea as an H-bond donor catalyst, thereby providing access to appealing cyclic hydrazino phosphonates and their derivatives. Mechanistic investigations suggest that the low solubility of these salts might favor the formation of a supramolecular thiourea-chloride-iminium 2:1:1 complex, which is proposed as the catalytically relevant species accounting for the observed nonlinear effect.
