Platelets assume a pivotal role in the cardiovascular diseases (CVDs). Thus, targeting platelet activation is imperative for mitigating CVDs. Ginkgetin (GK), from Ginkgo biloba L, renowned for its anticancer and neuroprotective properties, remains unexplored concerning its impact on platelet activation, particularly in humans. In this investigation, we delved into the intricate mechanisms through which GK influences human platelets. At low concentrations (0.5-1 μM), GK exhibited robust inhibition of collagen and arachidonic acid (AA)-induced platelet aggregation. Intriguingly, thrombin and U46619 remained impervious to GK's influence. GK's modulatory effect extended to ATP release, P-selectin expression, intracellular calcium ([Ca ]i) levels and thromboxane A formation. It significantly curtailed the activation of various signaling cascades, encompassing phospholipase Cγ2 (PLCγ2)/protein kinase C (PKC), phosphoinositide 3-kinase/Akt/glycogen synthase kinase-3β and mitogen-activated protein kinases. GK's antiplatelet effect was not reversed by SQ22536 (an adenylate cyclase inhibitor) or ODQ (a guanylate cyclase inhibitor), and GK had no effect on the phosphorylation of vasodilator-stimulated phosphoprotein or . Moreover, neither cyclic AMP nor cyclic GMP levels were significantly increased after GK treatment. In mouse studies, GK notably extended occlusion time in mesenteric vessels, while sparing bleeding time. In conclusion, GK's profound impact on platelet activation, achieved through inhibiting PLCγ2-PKC cascade, culminates in the suppression of downstream signaling and, ultimately, the inhibition of platelet aggregation. These findings underscore the promising therapeutic potential of GK in the CVDs.

Background And Objectives: The US Food and Drug Administration guidance for cold-stored platelets (CSPs) permits storage of apheresis platelets at 1-6°C for ≤14 days. During a pilot programme, CSPs were evaluated in a large US blood centre over a 10-month period (September 2023 to July 2024) to better understand the formation of aggregates under routine use.
Materials And Methods: Platelets collected in 100% plasma were moved into cold storage within 4 h of collection and shipped to local hospitals.

Platelet integrin αIIbβ3 is the final common effector of arterial thrombosis: it switches from a low-affinity to a high-affinity state, binds fibrinogen, and initiates the outside-in signals that stabilize a growing clot. Calcium- and integrin-binding protein 1 (CIB1) emerged as the first endogenous partner of the αIIb cytoplasmic tail and is now recognized as a dual-role adaptor. At rest, Ca-free CIB1 tethers the inner membrane clasp and restrains premature integrin activation; after ligand engagement, Ca-bound CIB1 docks onto αIIb, recruits focal-adhesion kinase and amplifies Src-dependent cytoskeletal remodeling.

Essential thrombocythemia (ET) is a myeloproliferative neoplasm (MPN) characterized by abnormal megakaryocyte proliferation and a markedly elevated platelet count, which predisposes patients to thrombotic or hemorrhagic events. Approximately 50%-60% of ET patients harbor a JAK2 V617F mutation. This mutation drives constitutive JAK kinase activation, promoting megakaryocyte proliferation and platelet production, while potentially activating inflammatory pathways and damaging vascular endothelium.
