Article Abstract

Nutritional strategies are required to limit the prevalence of denutrition in the elderly. With this in mind, fortified meals can provide more protein, but their digestibility must be ensured. Using a dynamic digester, DIDGI®, programmed with the digestion conditions of the elderly, we evaluated the supplementation of each component of a meal and assessed protein digestibility, amino acid profile, micro-nutrients and vitamins bioaccessibility for a full course meal. Higher protein digestibility was evidenced for the fortified meal, with higher release of essential amino acids. Moreover the large increase of leucine released was comparable to the range advocated for the elderly to favour protein anabolism. This study underlines the interest of using dish formulations to meet the nutritional needs of seniors, which is why this work will be completed by a clinical study in nursing home.

Mouse intestine as a useful model for CFTR electrophysiology function analysis.

Cystic fibrosis (CF) is a genetic disorder primarily known for its severe impact on lung function, but it also significantly affects the digestive system, leading to complications such as intestinal blockages, malabsorption, inflammation, and microbial dysbiosis. The study of CFTR (Cystic Fibrosis Transmembrane Conductance Regulator) effects on intestinal physiology is critical for developing new effective treatments. This work highlights the use of the mouse intestine as a valuable model for analyzing cellular electrophysiology and CFTR function.

Supercoiled (Sc) circular DNA, such as plasmids, are essential in molecular biology and hold strong therapeutic potential. However, they are typically produced in Escherichia coli, resulting in bacterial methylations, unnecessary sequences, and contaminants that hinder certain applications including clinical uses. These limitations could be avoided by synthesizing plasmids entirely in vitro, but synthesizing high-purity Sc circular DNA biochemically remains a significant technical challenge.

Parvalbumin (PV), a thermostable and digestion-resistant fish allergen, has been shown to retain its allergenic potential following traditional treatments, thus posing a persistent allergic risk. The study investigated the digestive kinetics and IgE immunoreactivity of Trachinotus ovatus PV, a major fish allergen, under different treatments (untreated; DPCD treatment-15 MPa, 30 min, 50 °C; heat treatment), to evaluate its allergenic potential alterations. The analysis was conducted using a combination of techniques to assess the proteolytic stability and IgE-binding capacity of PV, including Tris-Tricine-SDS-PAGE, Western blot (WB), indirect enzyme-linked immunosorbent assay (ELISA), and free amino group quantification.

Background: Early diagnosis of cholangiocarcinoma (CCA) remains challenging, but liquid biopsy is emerging as a promising detection strategy. Here, we identified a novel bile biomarker for CCA and developed an optic fiber biosensor integrated with digestive endoscopy for real-time diagnosis in vivo.

Methods: A total of 583 subjects and two proteomic analyses were used to screen and validate biomarkers for CCA, and then the corresponding antibodies were generated to construct a surface plasmon resonance (SPR)-based optic fiber biosensor.

The small intestine coordinates nutrient absorption and immune defense, but the epithelial signaling mechanisms bridging these functions are unclear. In this issue of Immunity, Yu et al. reveal a gasdermin-D-driven circuit that links lipid uptake and enterocyte metabolism to γδ T cell maintenance and barrier protection.
