Depression outcomes correlated to exposure to per- and poly-fluoroalkyl substances (PFAS).

Environ Sci Pollut Res Int

September 2025

Data Forward Analytics, LLC, Principal, Las Cruces, NM, 88011, USA.

Per- and poly-fluoroalkyl substances (PFAS) are fluorinated organic compounds known as forever chemicals that are used in many materials, from cooking appliances to industrial detergents. Given their high chemical stability due to their lipophilic properties and widespread use, PFAS tend to bioaccumulate in the cardiac, hepatic, renal, gastrointestinal, and neural tissues, resulting in a variety of malignant and non-malignant diseases. Depression is a prevalent mental health condition that affects an individual's activities of daily living.

This study investigated the effects of two emerging PFAS compounds, perfluorododecane sulfonic acid (PFDoDS) and perfluoro-4-ethylcyclohexane sulfonic acid (PFECHS), alongside legacy perfluorooctanesulfonic acid (PFOS), on gene expression in the liver, heart, and bursa of Fabricius from mallard ducklings (Anas platyrhynchos) exposed in ovo, simulating maternal transfer to the egg. These PFAS compounds were selected based on their detection in a declining sea duck species and concerns over their endocrine disruption potential. Farmed mallard eggs were injected with 80 ng/g of PFDoDS, PFECHS, or PFOS, reflecting concentrations at the upper end of those reported in wild bird eggs.

Aims: The aim of this research is to explore the suspected adverse drug reactions (ADRs) of perfluorinated medicines to determine whether side effects commonly associated with per- and poly-fluoroalkyl substances (PFAS) exposure were correlated to the type or number of fluorine atoms in these medications.

Methods: Thirteen fluorinated drugs and six non-fluorinated (or low fluorinated) comparators were selected after systematic triage. The reported ADR data from the Medicines and Healthcare Products Regulatory Agency's (MHRA) Yellow Card, and prescribing data from the OpenPrescribing database and the National Health Service Business Service Authority (NHSBSA) over a 5-year period were curated.

Metabolome informs about the chemical exposome and links to brain health.

Environ Int

Department of Psychiatry and Behavioral Sciences, Duke University, Durham, NC, USA; Department of Medicine, Duke University, Durham, NC, USA; Duke Institute of Brain Sciences, Duke University, Durham, NC, USA. Electronic address:

The metabolome is an intermediate phenotype, summarizing the profile of all small molecules (<1.5 kDa) in biospecimens. The metabolome provides a readout for the net influence of the chemical exposome, diet, gut microbiome, and genome on human health.

The Stream Pollution Trends Monitoring Program (SPoT) is one of several statewide programs under California's Surface Water Ambient Monitoring Program (SWAMP). SPoT has been generating sediment toxicity and pollutant data since 2008 from up to 100 diverse watersheds. Analysis has included toxicity testing with the amphipod Hyalella azteca and, more recently, the midge Chironomus dilutus, as well as the measurement of metals, polycyclic aromatic hydrocarbons (PAHs), polychlorinated biphenyls (PCBs), legacy pesticides, current use pesticides, and emerging contaminants such as fipronil, polybrominated diphenyl ethers (PBDEs) and per- and poly-fluoroalkyl substances (PFAS) in watershed sediments.
