AMPA receptors are members of the glutamate receptor family and mediate a fast component of excitatory synaptic transmission at virtually all central synapses. Thus, their functional characteristics are a critical determinant of brain function. We evaluate intolerance of each GRIA gene to genetic variation using 3DMTR and report here the functional consequences of 52 missense variants in GRIA1-4 identified in patients with various neurological disorders. These variants produce changes in agonist EC, response time course, desensitization, and/or receptor surface expression. We predict that these functional and localization changes will have important consequences for circuit function, and therefore likely contribute to the patients' clinical phenotype. We evaluated the sensitivity of variant receptors to AMPAR-selective modulators including FDA-approved drugs to explore potential targeted therapeutic options.

Diabetes mellitus (DM) is a chronic metabolic disorder characterized by persistent hyperglycemia with multiple clinical manifestations and complications, such as cardiovascular disease, kidney dysfunction, retinal impairment, and peripheral neuropathy. Continuous and minimally invasive glucose monitoring is essential for effective DM management. Microneedles (MNs)-based sensing platforms offer a promising solution; however, conventional polymeric MNs suffer from limited electrochemical sensitivity due to their insufficient electroactive surface area and inefficient loading of catalytic and enzymatic components.

Background: The cerebral circulation is continuously challenged by intravascular micrometer-sized particles that become trapped microvascular-emboli. These particles may include micro-thrombi, stiffened erythrocytes, and leukocytes, while also fat particles, air, and microplastics may cause microvascular embolism.
Review Scope: In this narrative review, we discuss these embolization processes and their acute and chronic consequences.

Introduction: Mutations in SORL1, encoding the sorting receptor Sortilin-related receptor with A-type repeats (SORLA), are found in individuals with Alzheimer's disease (AD). We studied SORLA, carrying a mutation in its ligand binding domain, to learn more about receptor functions relevant for human brain health.
Methods: We investigated consequences of SORLA expression in induced pluripotent stem cell (iPSC)-derived human neurons and microglia, using unbiased proteome screens and functional cell assays.
