Natural compounds are rich in pharmacological properties that are a hot topic in pharmaceutical research. The quinoline ring plays important roles in many biological processes in heterocycles. Many pharmacological compounds, including saquinavir and chloroquine, have been marketed as quinoline molecules with good anti-viral and anti-parasitic properties. Therefore, in this review, we summarize the medicinal chemistry of quinoline-modified natural product quinoline derivatives that were developed by several research teams in the past 10 years and find that these compounds have inhibitory effects on bacteria, viruses, parasites, inflammation, cancer, Alzheimer's disease, and others.

In single crystals of C-N atropisomeric -(2-halophenyl)quinolin-2-one and the thione analogue, a unique association based on a halogen-π interaction was detected. In racemic and optically pure -(2-bromo- or 2-chlorophenyl)quinolin-2-ones, homochiral layered polymers, which consist of ()- or ()-atropisomers, were formed through intermolecular halogen-π association. The halogen-π association in the racemates is due to a halogen bond (C-X···π) between a σ-hole on the halogen atom and a π-electron on the quinolinone benzene ring, while that in optically pure forms is caused by an n-π* interaction between a lone electron pair on the halogen atom and a π* orbital of the quinolinone.

In this study, a novel riboflavin-mediated nanoscale zero-valent iron/peracetic acid system (RF/nZVI/PAA) was constructed to increase the removal of norfloxacin. Under the optimal conditions (PAA=10 mg/L, nZVI=20 mg/L, RF= 1 mg/L, and initial pH =4), complete norfloxacin removal was achieved within 30 min, accompanied by a 70 % mineralization rate. Electron paramagnetic resonance spectroscopy combined with quenching experiments quantitatively identified hydroxyl radical, carbon-centered radical, and singlet oxygen as the predominant reactive oxidative species (ROS) responsible for norfloxacin removal, with contributions of 42 %, 44 %, and 10 %, respectively.

In refractory cases, traditional chemotherapy regimens and immunosuppressive drugs often prove ineffective. Artemisinin-based combination therapies (ACTs) remain the mainstay of treatment for malaria, despite reports of ACT treatment failure. ACTs consist of an artemisinin and a longer-lived partner drug, which is often a quinoline. Given that heme is central to the mechanism of action of artemisinins and some quinolines, we hypothesized that these antimalarials would exhibit strong drug-drug interactions.
