Alfaxalone increases measured progesterone concentration in neutered male cats when determined by immunoreactivity.

Article Abstract

Objective: Alfaxalone is a commonly used anesthetic agent in small animals. In cats, alfaxalone can be administered as an IM agent to achieve clinically useful sedation or anesthesia, negating the need for IV injection in difficult patients. The molecular structure of alfaxalone is similar to the hormone progesterone (P4). It is hypothesized that alfaxalone would cross-react with the assay measuring progesterone causing a false elevation.

Animals: 8 healthy neutered male, domestic shorthair cats that were privately owned were enrolled in the study.

Methods: Male neutered cats were administered 3 mg/kg of alfaxalone IM. Blood samples were collected at set time points (baseline, 30 minutes, 60 minutes, 3 hours, 6 hours, and 10 hours after administration), and serum concentrations of progesterone immunoreactivity (IR) were determined using the Siemens Immulite 1000 automated immunoassay system. Statistical analysis was performed with repeated measures ANOVA and a Tukey-Cramer multiple comparisons test. A P value of < .05 was used for significance.

Results: Serum progesterone IR was significantly elevated at 30 minutes, 1 hour, and 3 hours (P < .05) when compared to baseline progesterone immunoreactivity. Progesterone immunoreactivity had returned to baseline by 6 hours.

Clinical Relevance: This study suggests that alfaxalone administered IM in cats may interfere with immunoassay measurement of serum progesterone for up to 6 hours. Caution should be used when interpreting serum progesterone immunoreactivity results in cats within 4 hours of alfaxalone.

In this study, we evaluated the protective effects of combined melatonin and vitamin D3 treatment on ovarian reserve and tissue architecture in a cyclophosphamide-induced premature ovarian failure (POF) rat model. Forty-nine adult female rats were randomly assigned to seven groups, including intact control (group 1), single-agent control (groups 2 and 3), POF (group 4), and POF + treatment (groups 5, 6, and 7) groups. Cyclophosphamide exposure led to elevated FSH and LH levels, reduced estradiol and progesterone levels, extensive follicular atresia, stromal fibrosis, and the marked degeneration of the ovarian ultrastructure.

This report details findings from a morphologic, immunohistochemical, and molecular analysis of an example of endometrial mesonephric-like adenocarcinoma (MLA) with prominent clear cell carcinoma (CCC)-like features, with an assessment of whether such tumors represent mixed MLA/CCC or a morphologic subtype of MLA. Approximately 40% of an otherwise prototypical MLA was comprised of spatially discrete zones with prominent CCC-like morphology. The MLA-like nuclear features of both components were similar, as were their immunoreactivity patterns for estrogen receptor, progesterone receptor, SOX17, and calretinin (all negative), GATA3, and TTF1 (patchy positive).

Introduction: pelvic organ prolapse is a pathology of ligamentous connective tissues and matrix metalloproteinases are the main actors responsible for tissue remodeling by degrading connective tissue. The objective of this present study is to determine the expression rate of matrix metalloproteinases-1, -2 and -9 in prolapsed round ligaments and identify the types of matrix metalloproteinases most determining in the appearance of pelvic organs prolapse in in two pilot hospitals of Saint-Georges and of Bon-Berger of the city of Kananga in the Democratic Republic of Congo.

Methods: we conducted an analytical experimental study based on immunohistochemical examination of round ligaments of 100 consenting patients divided in two groups with and without pelvic organ prolapse treated in the gynecology departments of Bon-Berger hospitals in Tshikaji and Saint-Georges in Katoka in city of Kananga, from January 1 to July 31, 2023.
