Aims: Insomnia is a common sleep disorder, affecting up to 20% of the world population and adversely impacting productivity, health, and overall well-being. Although pharmacologic options exist to treat insomnia, the health-related quality of life for patients who are prescribed hypnotics is no higher than for those who are not, revealing a significant treatment gap. Sunobinop, an oral, selective, potent, partial agonist of the nociceptin receptor (NOP), was evaluated for efficacy and safety in the treatment of Diagnostic and Statistical Manual of Mental Disorders, Fifth Edition (DSM-5) insomnia disorder.

Background And Purpose: Migraine is a neurovascular disorder largely mediated by calcitonin gene-related peptide (CGRP). This study explores the role of the nociceptin/orphanin FQ (N/OFQ)-N/OFQ receptor (NOP) system in CGRP-induced periorbital mechanical allodynia (PMA) in mice.
Experimental Approach: Male or female wild type (NOP(+/+)) and NOP receptor knockout (NOP(-/-)) mice and CD-1 mice were used.

Migraine is a debilitating neurological disorder with significant impact on quality of life, including social functioning. This study investigated the effects of nitroglycerin (NTG)-induced migraine in male and female mice on social behavior and neuronal activation and explored the therapeutic potential of nociceptin opioid peptide (NOP) receptor agonism. We found that acute NTG administration induced mechanical allodynia in periorbital region and paw as well as impaired social behavior in both sexes, albeit with sex-specific patterns.

Rationale: A growing body of evidence suggests that the N/OFQ-NOP receptor system modulates learning and memory in rodents, with NOP receptor agonists impairing memory and antagonists reversing this effect. Moreover, previous studies on transgenic mice showed that genetic deletion of the NOP receptor enhances spatial and aversive memory.
Objectives: To further investigate the role of the NOP receptor system in learning and memory, we used a rat model to assess how genetic deletion of NOP receptors affects spatial and associative memory, comparing it with a wild-type (WT) control group.
Background: Nociceptin (N/OFQ) and its receptor OPRL1 play crucial roles in emotional processing, reward-related behaviors, learning, and neurotransmitter regulation, but their involvement in Alzheimer's disease (AD) pathology remains poorly understood. This study investigated possible relationships between prepronociceptin ( ) and expression and psychiatric symptoms in AD, utilizing Natural Language Processing (NLP) to assess Research Domain Criteria (RDoC) domains.
Methods: Post-mortem brain tissue from the dorsal anterior cingulate gyrus (BA32) was analyzed in 61 donors across different Braak and Braak (B&B) stages.
