Article Abstract

Products made from allogeneic tissue are largely used in clinical treatment due to its wide source compared with autologous tissue, causing less secondary trauma of patients and the good biocompatibility. Various organic solvents and other substances introduced in the production process of allogeneic products will leach down into the human through clinical treatment, thus bringing varying degrees of harm to patients. Therefore, it is very necessary to detect and control the leachables in such products. Based on the classification and summary of leachable substances existing in the allogeneic products, the preparation of extract and the establishment of the detection techniques for known and unknown leachable are briefly introduced in this study, in order to provide research method for the study of leachable substances of allogeneic products.

Drug product leachables are substances that are leached from the drug products manufacturing system components during manufacturing operations, drug product packaging systems during storage over shelf-life, and delivery systems during administration. Medical device leachables are substances that are released from medical devices during their clinical use. Thus, packaged drug products and medical devices are profiled for leachables (and/or extractables as probable leachables) to establish that the levels of leachables are sufficiently small that they present a negligible risk of adversely affecting patient health.

In this study, we propose a new metric for substance mobility in extension to the already implemented use of the log Koc as a sorption constant to the organic carbon content of soils. The new metric is based on the leachability of a compound in soil, ie the fraction thereof that is predicted to be transported to shallow groundwater. Apart from including climate and soil properties of European regions, the degradation half-life in soil is used in addition to the log Koc as an important parameter.
