Ischemia stroke is thought to be one of the vascular risks associated with neurodegenerative diseases, such as Alzheimer's disease (AD). Hydroxysafflor yellow A (HSYA) has been reported to protect against stroke and AD, while the underlying mechanism remains unclear. In this study, SH-SY5Y cell model treated with oxygen-glucose deprivation/reperfusion (OGD/R) was used to explore the potential mechanism of HSYA. Results from cell counting kit-8 (CCK-8) showed that 10 μM HSYA restored the cell viability after OGD 2 hours/R 24 hours. HSYA reduced the levels of malondialdehyde and reactive oxygen species, while improved the levels of superoxide dismutase and glutathione peroxidase. Furthermore, apoptosis was inhibited, and the expression of brain-derived neurotrophic factor was improved after HSYA treatment. In addition, the expression levels of amyloid-β peptides (Aβ) and BACE1 were decreased by HSYA, as well as the expression levels of binding immunoglobulin heavy chain protein, PKR-like endoplasmic reticulum (ER) kinase pathway, and activating transcription factor 6 pathway, whereas the expression level of protein disulfide isomerase was increased. Based on these results, HSYA might reduce Aβ toxicity after OGD/R by interfering with apoptosis, oxidation, and neurotrophic factors, as well as relieving ER stress.

This study aims to explore the effects and action mechanisms of the active ingredients in Buyang Huanwu Decoction(BYHWD), namely tetramethylpyrazine(TMP) and hydroxy-safflor yellow A(HSYA), on oxygen-glucose deprivation/reglucose-reoxygenation(OGD/R)-induced inflammation and oxidative stress of microglia(MG). Network pharmacology was used to screen the effective monomer ingredients of BYHWD and determine the safe concentration range for each component. Inflammation and oxidative stress models were established to further screen the best ingredient combination and optimal concentration ratio with the most effective anti-inflammatory and antioxidant effects.

Osteoporosis is hallmarked by marrow adiposity, whereas the involvement of sphingosine kinase-1(SphK1)/sphingosine 1-phosphate (S1P)/sphingosine 1-phosphate receptors(S1PR) mediated signaling in adipocyte/osteoblast lineage commitment remains elusive. While Hydroxysafflor yellow A (HSYA) attenuates estrogen deficiency-induced bone loss, its pharmacological mechanisms remain incompletely elucidated. Our investigations in ovariectomized (OVX) murine models revealed that SphK1 ablation diminished osteoblast-specific markers (Procollagen type I N-terminal propeptide [PINP], Osteocalcin [OCN], Osteoprotegerin [OPG]), disrupted trabecular microarchitecture, and exacerbated adipose conversion through suppression of SphK1/S1PR2 coupled with Peroxisome proliferator-activated receptor gamma (PPARγ) upregulation.

Ethnopharmacological Relevance: Buyang Huanwu Decoction (BYHWD) is traditionally prescribed for Qi-deficiency and blood-stasis stroke in TCM, yet its key pharmacologically active constituents and mechanistic basis remain to be fully elucidated.
Aim Of The Study: To investigate the key pharmacologically active constituents of BYHWD against Qi-deficiency and blood-stasis type cerebral ischemia-reperfusion injury (QDBS + CI/R), and to clarify their roles in counteracting oxidative stress, suppressing inflammation, and improving energy metabolism.
Methods: A QDBS + CI/R rat model was used to assess BYHWD's therapeutic effects via neurological and syndrome scoring, histology (HE, Nissl), and biochemical markers (T-AOC, IL-1β, IL-6, ATP, ADP).
