An immunoaffinity layer with orientation-controlled antibodies was constructed to express streptococcal protein G in cells using autodisplay technology. The sequence of protein G, a specific IgG-binding protein, was inserted into the autodisplay vector using recombinant technology and the constructed plasmid vector was transformed into cells. Protein G was confirmed to be autodisplayed with a high density of 2 × 10 copies per cell by SDS-PAGE analysis, and its IgG-binding affinity was confirmed by fluorescence microscopy. Autodisplayed protein G showed higher affinity than the IgG-binding Z-domain for goat IgG. Immunoassays based on cells were established to detect horseradish peroxidase (HRP) and C-reactive protein (CRP). Protein G autodisplaying cells were utilized as a solid support and immunoassays showed improved sensitivity by orientation control of autodisplayed protein G. The outer membrane (OM) of protein G autodisplaying was isolated and layered to construct an immunoaffinity layer. The OM was coated on a microplate to perform the immunoassays, which showed limits of detection of 5 and 0.2 ng mL for HRP and CRP, respectively. An OM layer with autodisplayed protein G was applied as the immunoaffinity layer of a surface plasmon resonance (SPR) biosensor. After CRP detection, the SPR responses showed good linearity, with an value of 0.99. The immunoaffinity layer with orientation control by autodisplayed protein G was confirmed to be applicable in immunoassays and immunosensors to improve sensitivity.

AMPA-type ionotropic glutamate receptors (AMPARs) are integral to fast excitatory synaptic transmission and have vital roles in synaptic plasticity, motor coordination, learning and memory. Whereas extensive structural studies have been conducted on recombinant AMPARs and native calcium-impermeable (CI)-AMPARs alongside their auxiliary proteins, the molecular architecture of native calcium-permeable (CP)-AMPARs has remained undefined. Here, to determine the subunit composition, physiological architecture and gating mechanisms of CP-AMPARs, we visualize these receptors, immunoaffinity purified from rat cerebella, and resolve their structures using cryo-electron microscopy (cryo-EM).

Fetomaternal hemorrhage, characterized by the passage of fetal blood into maternal circulation, poses significant risks to both mother and fetus. As it may be diagnosed by quantifying fetal red blood cells in maternal circulation, we developed an immunosensor to detect fetal cells made with a layer-by-layer silk fibroin film and a commercial CD71 monoclonal antibody. Detection was achieved through electrical impedance spectroscopy, allowing discrimination between human umbilical cord blood (UCB) cells and adult peripheral blood (APB) cells from patients and volunteers.

Silk was the driving force behind the opening of the Silk Road, positioning this material as a global commodity that greatly influenced the progress of human civilization. Due to the natural protein properties of silk, the internal structure and external characteristics of silk cultural relics are irreversibly destroyed during the process of burial or when passed down through the generations until the production information and material components completely disappear, making it difficult to obtain direct archaeological evidence for pinpointing the origin of silk. The sacrificial pits at the Sanxingdui archaeologcal site, located in Guanghan, Sichuan, China, have been found with layers of ash above the layers of artifacts and some bronzes with fabric traces.
