The continuing rise in the atmospheric carbon dioxide (CO) concentration causes stomatal closing, thus critically affecting transpirational water loss, photosynthesis, and plant growth. However, the primary CO sensor remains unknown. Here, we show that elevated CO triggers interaction of the MAP kinases MPK4/MPK12 with the HT1 protein kinase, thus inhibiting HT1 kinase activity. At low CO, HT1 phosphorylates and activates the downstream negatively regulating CBC1 kinase. Physiologically relevant HT1-mediated phosphorylation sites in CBC1 are identified. In a genetic screen, we identify dominant active HT1 mutants that cause insensitivity to elevated CO. Dominant HT1 mutants abrogate the CO/bicarbonate-induced MPK4/12-HT1 interaction and HT1 inhibition, which may be explained by a structural AlphaFold2- and Gaussian-accelerated dynamics-generated model. Unexpectedly, MAP kinase activity is not required for CO sensor function and CO-triggered HT1 inhibition and stomatal closing. The presented findings reveal that MPK4/12 and HT1 together constitute the long-sought primary stomatal CO/bicarbonate sensor upstream of the CBC1 kinase in plants.

Rapid-acting antidepressants (RAADs) such as ketamine are currently under development. The aim of this study is to characterize the neural circuits affected by ketamine and NLX-101, a selective 5-HT receptor biased agonist which has shown promising effects, by using [F]FDG PET imaging in rats that had received chronic administration of corticosterone (CORT), a model of anxiety-depression. In a longitudinal study, regional changes in brain activity were investigated in 24 selected CORT rats.

Background: Aneurysmal subarachnoid hemorrhage (aSAH) is notoriously known for its high mortality and morbidity. Quadrupolar nuclei with a nuclear spin I > 1/2 account for ∼73 % of all NMR-active nuclei. The quadrupolar interaction broadens solid-state NMR spectra, frequently resulting in low resolution and poor sensitivity. Through its widespread reciprocal connections with the cerebral cortex, the claustrum is implicated in sleep and waking cortical network states. Yet, basic knowledge of neuromodulation in this structure is lacking. The claustrum is richly innervated by serotonergic fibers, expresses serotonin receptors, and is suggested to play a role in the ability of psilocybin, which is metabolized to the non-specific serotonin receptor agonist psilocin, to disrupt cortex-wide network states.
