Ordered supramolecular hydrogels assembled by modified aromatic amino acids often exhibit low mechanical rigidity. Aiming to stabilize the hydrogel and understand the impact of conformational freedom and hydrophobicity on the self-assembly process, two building blocks based on 9-fluorenyl-methoxycarbonyl-phenylalanine (Fmoc-Phe) gelator which contain two extra methylene units in the backbone, generating Fmoc-γPhe and Fmoc-(3-hydroxy)-γPhe are designed. Fmoc-γPhe spontaneously assembled in aqueous media forming a hydrogel with exceptional mechanical and thermal stability. Moreover, Fmoc-(3-hydroxy)-γPhe, with an extra backbone hydroxyl group decreasing its hydrophobicity while maintaining some molecular flexibility, self-assembled into a transient fibrillar hydrogel, that later formed microcrystalline aggregates through a phase transition. Molecular dynamics simulations and single crystal X-ray analyses reveal the mechanism underlying the two residues' distinct self-assembly behaviors. Finally, Fmoc-γPhe and Fmoc-(3-OH)-γPhe co-assembly to form a supramolecular hydrogel with notable mechanical properties are demonstrated. It has been believed that the understanding of the structure-assembly relationship will enable the design of new functional amino acid-based hydrogels.

Elaborate kinetic control enables supramolecular self-assemblies to deviate from equilibrium, depicting profound energy landscapes with remarkable structural and functional diversity. Despite this potential, achieving an energy landscape that encompasses lamellar and inverse cubic structures remains a significant challenge, contrasting with the sophisticated structural transformations naturally orchestrated by cellular systems. Here, we present a dynamic and minimalistic Janus dendrimer self-assembly system capable of reversibly transitioning between lamellar vesicles and inverse cubic structures.

Molecular recognition is essential for controlling chemical processes, passing molecular instructions to elicit responses including structure formation, signalling and replication. Usually, the selectivity of molecular recognition is under thermodynamic control; however, when a higher fidelity is required, nature improves recognition selectivity by an error correction mechanism under an energy-dissipating kinetic-control regime. Here, exploiting DNA hybridization as a model, we present an abiotic example of an information ratchet mechanism that increases selectivity for the 'correct' duplex from 2:1 at equilibrium to 6:1 under energy-dissipating conditions.

Despite the multiple roles of flexible and achiral Gly in regulating protein architectures and functions, its high flexibility is seldom exploited as a structural modulator in the design of self-assembling peptides. By using minimalistic peptide sequences, the effects of Gly insertions are investigated on the molecular conformation and the supramolecular morphology, focusing on Gly-induced bent structures and their impact on self-assembled nanostructures and handedness. Different backbone bending degrees are generated by varying Gly position, which in turn resulted in distinct hydrogen bonding modes and residue shifting upon dimerization, eventually leading to β-sheets and nanofibrils with opposite handedness.
