Current biopharmaceutical drugs are mainly a class of peptides or proteins that play an essential role in the treatment of many diseases. Such peptides/proteins are usually thermally unstable and may lose their bioactivity when exposed to ambient conditions. Therefore, they are not suitable for long-term storage. Lyophilisation is the most common method to prolong shelf life of solid peptide/protein drugs; however, the freeze-drying process can lead to irreversible damage. In the present study, human interferon-alpha 2a (IFN-α2a) was selected as a model protein drug; four disaccharides (β-lactose, β-maltose, sucrose, and trehalose) were selected as bioactive protectants. We investigated the effects of different protectants on IFN-α2a under various ambient conditions (vacuum, dry state, and aqueous solution) using replica exchange molecular dynamics simulation. The protective effect of β-maltose on IFN-α2a was the highest in aqueous solution and dry state, β-lactose showed a poor protective effect in all three conditions, the performance of sucrose was good in all conditions, and trehalose showed a better protective effect under vacuum conditions and in aqueous solution. Disaccharides form H-bonds with water, thereby preventing water from the tertiary structure of proteins. Trehalose forms strong H-bonds with water which explains its extraordinary stability.

Accurately modeling volume-dependent properties of water remains a challenge for density functional theory (DFT), with widely used functionals failing to reproduce key features of the water density isobar, including its shape, density, and temperature of the density maximum. Here, we compare the performance of the RPBE-D3 and vdW-DF-cx functionals using replica exchange molecular dynamics (MD) driven by machine-learned force fields. Our simulations reveal that vdW-DF-cx predicts the water density more accurately than RPBE-D3 and reproduces the isobar closely between 307 and 340 K.

Parkinson's disease (PD) is significantly characterized by the accumulation of α-synuclein (α-Syn) amyloid aggregates, especially in the form of Lewy bodies. Our study explores the effect of one of the four nucleobases, adenine, on the amyloid transformation of the A53T mutant of α-Syn (A53T Syn), which is linked to the early-onset PD characterized by increased protofibril production and fast aggregation. Systematic analysis using biophysical techniques in conjunction with computational methods demonstrated that adenine stabilizes the monomeric conformations of A53T Syn by interacting with the NAC domain of the protein through non-covalent interactions.

Cationic antimicrobial peptides (AMPs) are toxic to microbes, such as bacteria and fungi, and have been increasingly studied as an alternative to traditional antibiotics, in part because AMPs are bactericidal with a minimum risk of developing bacterial resistance. Indolicidin (IL) is an AMP derived from bovine neutrophils that is unique due to its high prevalence of tryptophan and proline amino acids and its disordered structure. In addition to its antimicrobial activity, IL has exhibited toxicity toward mammalian cells, resulting in hemolysis.

Ca-ATPases in the plasma membrane extrude Ca ions from the cytosol to the extracellular space thereby terminating Ca-signals and controlling Ca-homeostasis in any type of cell. Recently, these Ca-pumps have been identified as protein complexes of the transporting subunits PMCAs1-4 and the single-span membrane proteins Neuroplastin (NPTN) or Basigin that are obligatory for efficient trafficking of the pump complexes to the surface membrane. Quantitative investigation of the pumping velocity controlling the time course of Ca-signals, however, has remained unresolved.
