Background: Ovarian cancer could induce alterations in both structure and function of the brain. This study employs Mendelian randomization (MR) to investigate the causal relationship between brain imaging-derived phenotypes (IDPs) and ovarian cancer, offering new insights into the potential clinical applications of IDPs for ovarian cancer risk assessment.

Imaging transcriptomics has become a power tool for linking imaging-derived phenotypes (IDPs) to genomic mechanisms. Yet, its potential for guiding CNS drug discovery remains underexplored. Here, utilizing spatially-dense representations of the human brain transcriptome, we present an analytical framework for the transcriptomic decoding of high-resolution surface-based neuroimaging patterns, and for linking IDPs to the transcriptomic landscape of complex neurotransmission systems in vivo.

Background And Aim: Existing polygenic risk scores (PRS) for severe liver disease (SLD) have limited predictive ability, highlighting a possible reorientation for PRS application in clinical practice.

Methods: Using non-overlapping subsets of the UK Biobank cohort, we first conducted a genome-wide association study of magnetic resonance imaging-derived hepatic fat content (HFC; n = 12,838), and then constructed a polygenic risk score to capture genetically predicted HFC (gHFC), which was applied in an independent sample (n = 426,529) to stratify individuals and evaluate the performance of clinical fibrosis scores.

Results: Among 426,529 participants, 4417 developed SLD during follow-up.
