Article Abstract

Atopic dermatitis (AD) is a chronic inflammatory skin disease characterized by barrier dysfunction, dysregulated immune response, and dysbiosis with increased colonization. Infiltration of various T helper cell subsets into lesional skin and subsequent cytokine release are a hallmark of AD. Release of cytokines by both T cells and keratinocytes plays a key role in skin inflammation and drives many AD features. This review aims to discuss cytokine-mediated crosstalk between T cells and keratinocytes in AD pathogenesis and the potential impact of virulence factors produced by on these interactions.

BDNF as a Key Mediator in Eosinophilic Inflammation in CRSwNP: Insights From Transcriptomic and Functional Analysis.

Department of Otolaryngology Head and Neck Surgery and Department of Allergy, Beijing TongRen Hospital, Capital Medical University, Beijing, China.

Background: Chronic rhinosinusitis with nasal polyps (CRSwNP) is a heterogeneous inflammatory disorder characterized by eosinophil-driven pathology. While neurotrophins, particularly brain-derived neurotrophic factor (BDNF), have been implicated in eosinophilic inflammation in allergic conditions, their involvement in CRSwNP remains undefined.

Methods: Bulk-RNA sequencing of 23 nasal mucosa samples and ELISA quantification of 95 nasal secretions were conducted to evaluate BDNF expression across CRSwNP endotypes and its correlation with eosinophil infiltration and clinical severity.

Tumor stroma, or tumor microenvironment (TME), has been in the spotlight during recent years for its role in tumor development, growth, and metastasis. It consists of a myriad of elements, including tumor-associated macrophages, cancer-associated fibroblasts, a deregulated extracellular matrix, endothelial cells, and vascular vessels. The release of proinflammatory molecules, due to the inflamed microenvironment, such as cytokines and chemokines is found to play a pivotal role in progression of cancer and response to therapy.

Background: The mechanisms shared and unique to thrombotic diseases (TDs) remain unclear. This study explores the causal relationships between inflammatory cytokines and TDs via multi-omics integration to identify targeted therapeutic alternatives.

Methods: We conducted two-sample Mendelian randomization (MR) using three genetic instruments to evaluate the causal effects of 47 cytokines on 13 TD traits.

Study Question: Does leukocyte-derived Oncostatin-M (OSM) regulate the ovulatory process in human dominant follicles?

Summary Answer: Leukocyte-derived OSM activates key signaling pathways in human preovulatory granulosa cells and modulates steroidogenesis, prostaglandin synthesis, and tissue remodeling in human ovulatory follicles.

What Is Known Already: Leukocytes are essential regulators of ovulation. Our recent single-cell RNA sequencing (scRNA-seq) has identified diverse leukocyte subpopulations in follicular aspirates obtained from IVF patients and revealed the expression of OSM in leukocytes and its receptors (OSMR, LIFR, IL6ST) in follicular cells.
