The gene regulation underlying axon formation and its exclusiveness to neurons remains elusive. TRIM46 is postulated to determine axonal fate. We show Trim46 mRNA is expressed before axonogenesis, but TRIM46 protein level is inhibited by alternative splicing of two cassette exons coupled separately to stability controls of Trim46 mRNA and proteins, effectively inducing functional knockout of TRIM46 proteins. Exon 8 inclusion causes nonsense-mediated mRNA decay of Trim46 transcripts. PTBP2-mediated exon 10 skipping produces transcripts encoding unstable TRIM46 proteins. During axonogenesis, transcriptional activation, decreased exon 8 inclusion, and enhanced exon 10 inclusion converge to increase TRIM46 proteins, leading to its neural-specific expression. Genetic deletion of these exons alters TRIM46 protein levels and shows TRIM46 is instructive though not always required for AnkG localization nor a determinant of AnkG density. Therefore, two concurrently but independently regulated alternative exons orchestrate the temporal induction and tissue-specific expression of TRIM46 proteins to mediate axon formation.

Milk protein percentage (PP) and fat percentage (FP) are important indicators for measuring milk quality, but only a few causative genes such as DGAT1, GHR, and ABCG2, have been identified, indicating substantial potential for further exploration. Here, we integrated genotyping, RNA-seq, ATAC-seq, single-cell atlas and cis-QTLs datasets from the liver and mammary gland to investigate their genetic regulation. We identified cell type affecting milk composition, such as HPE2 and LumSec-HSPH1, and highlighted nine candidate genes (EFNA1, ERBB3, DUSP16, DEPTOR, TRIM46, HSTN, CIDEA, ACACA and SPP1) that are involved in the regulation of milk protein and fat synthesis through MAPK, AMPK, PI3K-Akt, and mTOR signalling pathways.

Microtubules (MTs) in the axon are nearly all oriented with plus-end-out, and this pattern of organization is important for regulating the morphology and cytoplasmic composition of the axon, and for defining the cargoes transported in the anterograde and retrograde directions in the axon. Over the past several years, studies on vertebrate neurons (and also insect neurons) have been conducted by many investigators to understand the mechanisms that establish this pattern and maintain it over the life of the neuron and in the face of potential corruption. Studies on cultured rat sympathetic neurons from superior cervical ganglia (and also insect neurons) have implicated sliding of MTs by cytoplasmic dynein as crucial not only for establishing the plus-end-out orientation of axonal MTs but also for clearing mal-oriented MTs from the axon.
