Background And Objective: AMG 986 is a first-in-class, novel apelin receptor small molecule agonist initially developed for the treatment of heart failure. The current phase I study was conducted to evaluate the pharmacokinetics and safety of a single-dose 200-mg capsule formulation of AMG 986 relative to the tablet formulation in 12 healthy subjects.
Methods: In a two-period, two-way crossover design, eligible subjects were randomized 1:1 to tablet/capsule or capsule/tablet treatment sequences; each treatment sequence lasted for approximately 6 days and comprised six subjects.
Results: Following a single oral dose of AMG 986, the geometric mean maximum observed concentration (C) values were 9670 ng/mL and 6920 ng/mL and the geometric mean area under the curve from time zero to 120 h (AUC) values were 68,000 ng*h/mL and 59,900 ng*h/mL for the tablet and capsule, respectively. The geometric least squares means (90% confidence interval [90% CI]) for the ratios of capsule/tablet were 0.88 (90% CI 0.81-0.96) and 0.72 (90% CI 0.57-0.91) for AUC and C, respectively. AMG 986 had an acceptable safety profile; all adverse events were grade 1 or 2 in severity.
Conclusion: There was a modest 12% decrease in AUC and a 28% decrease in C with the AMG 986 capsule versus the tablet. These differences are not considered to be clinically relevant, suggesting the capsule formulation can be used in subsequent clinical studies of AMG 986.

Objective: CXCR4 and integrin αβ play important roles in tumor biology and are highly expressed in multiple types of tumors. This study aimed to synthesize, preclinically evaluate, and clinically validate a novel dual-targeted PET imaging probe Ga-pentixafor-c(RGDfK) for its potential in imaging tumors.
Methods: The effects of Ga-pentixafor-c(RGDfK) on cell viability, targeting specificity, and affinity were assessed in the U87MG cells.

Tamsulosin is a highly selective α1A adrenergic receptor antagonist that can relax smooth muscles in the urethra, bladder neck, and prostate and improve urinary disorders. It is therefore widely used to treat lower urinary tract symptoms caused by benign prostatic hyperplasia. The aim of this study is to evaluate the pharmacokinetic (PK) characteristics and bioequivalence of 2 different formulations (tamsulosin sustained-release tablets and tamsulosin sustained-release capsules) in healthy Chinese subjects.

Topiramate is increasingly used in the treatment of epilepsy during pregnancy. However, its plasma concentration evidently decreases during pregnancy, which may reduce its efficacy. This study aimed to develop a physiologically based pharmacokinetic (PBPK) model of topiramate to simulate maternal and fetal pharmacokinetic changes across different trimesters and to propose dose adjustments.

The aim of this Phase 1, multicentre, open-label study was to evaluate the safety, tolerability and pharmacokinetics (PK) of abemaciclib administered at global recommended Phase 2 dose (RP2D) of 200 mg twice daily, combined with standard doses of abiraterone and prednisolone, in Japanese patients with metastatic castration-resistant prostate cancer (mCRPC). Dose-limiting toxicities (DLTs) were assessed for 28 days post-first dose. Six patients were treated, and all experienced at least one treatment-emergent adverse event (TEAE), mostly low grade; no Grade 4 or 5 TEAEs occurred.

The cardiovascular continuum is the developmental process of cardiovascular diseases (CVDs) leading to heart failure (HF) and sudden cardiac death. Beta-blockers (BBs) are at the forefront of managing conditions along this continuum, ranging from cardiovascular (CV) risk factors to heart failure. In particular, bisoprolol proved to be a highly cardio-selective BB with a favourable pharmacokinetic profile, demonstrating long-term safety, good tolerability, and proven efficacy in reducing cardiac events, including arrhythmias and mortality in patients with heart failure with reduced ejection fraction (HFrEF).
