Melanoma is a drug-resistant cancer, representing a serious challenge in cancer treatment. Dacarbazine (DTIC) is the standard drug in metastatic melanoma treatment, despite the poor results. Hyperthermia has been proven to potentiate chemotherapy. Hence, this work analyzed the combined action of hyperthermia and DTIC on A375 and MNT-1 cell lines. First, temperatures between 40 °C and 45 °C were tested. The effect of DTIC on cell viability was also investigated after exposures of 24, 48, and 72 h. Then, cells were exposed to 43 °C and to the respective DTIC IC10 or IC20 of each time exposure. Overall, hyperthermia reduced cell viability, however, 45 °C caused an excessive cell death (>90%). Combinational treatment revealed that hyperthermia potentiates DTIC’s effect, but it is dependent on the concentration and temperature used. Also, it has different mechanisms from the treatments alone, delaying A375 cells at the G2/M phase and MNT-1 cells at the S and G2/M phases. Intracellular reactive oxygen species (ROS) levels increased after treatment with hyperthermia, but the combined treatment showed no additional differences. Also, hyperthermia highly increased the number of A375 early apoptotic cells. These results suggest that combining hyperthermia and DTIC should be more explored to improve melanoma treatment.

Benign prostatic hyperplasia (BPH) presents a significant clinical challenge, with conventional therapies carrying substantial risks, including urinary retention, sexual dysfunction, and prolonged recovery. To address the urgent need for safer, ultra-minimally invasive alternatives, we developed a sonosensitizing nanoplatform using copper-manganese-doped mesoporous silica nanoparticles (Cu-Mn@SiO) for ultrasound-induced sonodynamic therapy (SDT). Here, we demonstrate that this innovative strategy provides highly effective and precisely targeted therapy for BPH.

Nanomedicine has developed an impressive array of anticancer devices, from drug delivery vectors to hyperthermia-enabling nanoparticles. Lately, emphasis has been shifted to the tumour microenvironment (TME) and to active nanomaterials adapted to the prevailing conditions. Among them, catalytic nanomaterials (also called nanocatalysts) represent a highly interesting alternative, especially when they are able to target metabolites that are present to a greater extent in tumoral cells, opening a window for a catalytically selective action.

This case report describes a unique instance of refractory paroxysmal sympathetic hyperactivity (PSH) in a 19-year-old woman following a traumatic brain injury sustained in a motorcycle accident. The patient presented in a semicomatose state with a Glasgow Coma Scale score of 3 (E1, VT, M2), a significant left frontotemporal subdural hematoma, and a midline shift that necessitated emergency craniectomy and hematoma evacuation. Postoperatively, she developed recurrent episodes of hyperthermia, tachycardia, hypertension, tachypnea, diaphoresis, rigidity, and eyeball deviation triggered by non-noxious stimuli.

Mild photothermal therapy effectively reduces hyperthermia-related tissue injuries; however, its low therapeutic efficiency presents a significant limitation. Herein, we developed an organic multifunctional photothermal platform with enhanced Fenton catalytic activity to increase tumor sensitivity to mild photothermal therapy. This platform incorporated a self-augmented Fenton molecule integrating carbonic anhydrase inhibitor and ferrocene, which was co-encapsulated with the photothermal agent IR 825 via an amphiphilic polymer.

Chronic periodontitis, a frequent complication of diabetes, is exacerbated by bacterial biofilms that drive progressive periodontal tissue destruction and systemic inflammation. Conventional treatments, utilizing mechanical debridement and systemic antibiotics, often fail to eradicate bacterial biofilms, promote antibiotic resistance, and lack real-time monitoring, leading to suboptimal therapeutic outcomes. Herein, we report a separable bilayer microneedle (MN) patch that enables localized, antibiotic-free, biofilm-targeted therapy and in situ biomarker-based monitoring for the integrated management of chronic periodontitis.
