Through regulation of the epigenome, the bromodomain and extra terminal (BET) family of proteins represent important therapeutic targets for the treatment of human disease. Through mimicking the endogenous -acetyl-lysine group and disrupting the protein-protein interaction between histone tails and the bromodomain, several small molecule pan-BET inhibitors have progressed to oncology clinical trials. This work describes the medicinal chemistry strategy and execution to deliver an orally bioavailable tetrahydroquinoline (THQ) pan-BET candidate. Critical to the success of this endeavor was a potency agnostic analysis of a data set of 1999 THQ BET inhibitors within the GSK collection which enabled identification of appropriate lipophilicity space to deliver compounds with a higher probability of desired oral candidate quality properties. SAR knowledge was leveraged via Free-Wilson analysis within this design space to identify a small group of targets which ultimately delivered I-BET567 (), a pan-BET candidate inhibitor that demonstrated efficacy in mouse models of oncology and inflammation.

During seed development, vacuolar sorting receptors (VSRs) recognize a sequence-specific vacuolar sorting determinant located at the C terminus (ctVSD) of storage proteins, thereby sorting them into protein storage vacuoles. The protease-associated (PA) domain of VSRs is responsible for interacting with the ctVSD of cargo proteins. Here, we report the crystal structure of the PA domain of Arabidopsis vacuolar-sorting receptor 1 (VSR1) in complex with the C-terminal pentapeptide (SDRFV) of vicilin-like seed storage protein 22 (VL22).

Second mitochondrial activator of caspase (SMAC) mimetics (SMACm) and bromodomain and extra-terminal domain (BET) inhibitors (BETi) are two distinct classes of novel anticancer therapeutics. So far, broad clinical benefit for either monotherapy has not been achieved, calling for effective combination strategies. We show that the combination of BI 891065, a monovalent oral SMACm antagonist of inhibitor of apoptosis protein 1 (cellular inhibitor of apoptosis protein 1 [cIAP1]), and BI 894999, a potent and selective oral BETi, significantly impaired cancer cell proliferation irrespective of tissue context.

Methods: This was a retrospective multicenter study of all patients with a thoracic, abdominal or pelvic tumor diagnosed prenatally or within the first 3 months after birth in the Île-de-France area between January 2010 and May 2021.

This study presents a hybrid modeling framework for predicting proppant settling rate (PSR) in hydraulic fracturing by integrating symbolic physics-based derivations, parametric simulations, and ensemble machine learning. Symbolic expressions were formulated using Stokes' law, drag equations, and pressure-gradient dynamics. A symbolic dataset was synthetically generated by sampling realistic physical ranges: proppant density [Formula: see text], fluid viscosity [Formula: see text], and particle diameter [Formula: see text].
