Article Abstract

Introduction: Mediastinal lymph node (MLN) removal by video-assisted mediastinoscopic lymphadenectomy (VAMLA) for preoperative cancer staging was reported to be associated with increased survival. The aim of this study was to evaluate the immunologic effects of complete MLN removal by VAMLA on cytotoxic T lymphocyte (CTL) phenotype and function.

Methods: Seventeen patients with non-small cell lung cancer (NSCLC) (stage cT1-4N0-3M0-1A) and 20 healthy participants were included in this study. Blood samples were collected before and 4 weeks after the procedure. Lymphocytes were isolated from the removed MLNs. CTL phenotypes and functions were evaluated by flow cytometry. Plasma levels of soluble programmed cell death protein 1 (sPD-1), soluble programmed cell death protein 1 ligand, and soluble CTL antigen 4 (sCTLA-4) were measured with enzyme-linked immunosorbent assay.

Results: The ratio of the immunosenescent CTLs (CD3CD8CD28) was increased in peripheral blood and MLNs of the patients with NSCLC compared to controls ( = 0.037), and MLN removal did not change this ratio. PD-1 and CTL antigen 4 expressions were significantly reduced in peripheral blood CTLs after MLN removal by VAMLA ( = 0.01 and = 0.01, respectively). Granzyme A expression was significantly reduced in the peripheral blood CTLs of the patients compared to controls ( = 0.006) and MLN removal by VAMLA significantly improved Granzyme A expression in CTLs ( = 0.003). Plasma concentrations of sPD-1 and sCTLA-4 remained unchanged after VAMLA.

Conclusion: CTLs in the MLNs and peripheral blood of the patients with NSCLC had an immunosenescent phenotype, increased immune checkpoint receptor expression, and impaired cytotoxicity. MLN removal by VAMLA improved these phenotypic and functional characteristics of CTLs. These changes may explain the potential contribution of VAMLA to improved survival.

Objective: To evaluate the reliability and effectiveness of using the radioactive iodine seed (RIS) as one-step target axillary dissection (TAD) procedure in breast cancer patients.

Materials And Methods: This was a non-randomized, single-center, prospective study. Consecutive patients between May-2022 and November-2023 with breast cancer and axillary involvement (cN1) planned to receive NAC and TAD surgery were marked with RIS in the pathologically proven ALN.

Magnetic lignin nanoparticles (MLNs) were prepared by inducing their self-assembly through lignin regeneration in the [N-methyl-2-pyrrolidone][C1-C4 carboxylic acid] ionic liquids ([NMP]ILs), which are low-cost protic ionic liquid. [NMP]ILs are self-assembling solvent that can enhance the adsorption capacity of MLNs to a greater degree than tetrahydrofuran or HO. Additionally, the anion types of [NMP]IL greatly influence the physiochemical properties of MLNs.

Article Synopsis
  • Surface-enhanced Raman spectroscopy (SERS) is a sensitive technique for detecting tiny amounts of substances, but the reliability of results is affected by the properties of the substrates used.
  • This study introduces a new, cost-effective nanocomposite made of silver nanoparticles and porous magnesium oxide, which enhances SERS performance due to its unique structure that creates more active sites for adsorption.
  • The nanocomposite effectively detects a variety of environmental pollutants at low concentrations with high stability and reproducibility, offering a promising tool for monitoring contaminants in different fields.

Male meiotic spindle poles are stabilized by TACC3 and cKAP5/chTOG differently from female meiotic or somatic mitotic spindles in mice.

Transforming acidic acid coiled-coil protein 3 (TACC3) and cytoskeleton associated protein 5 (cKAP5; or colonic hepatic tumor overexpressed gene, chTOG) are vital for spindle assembly and stabilization initiated through TACC3 Aurora-A kinase interaction. Here, TACC3 and cKAP5/chTOG localization with monospecific antibodies is investigated in eGFP-centrin-2- expressing mouse meiotic spermatocytes. Both proteins bind spermatocyte spindle poles but neither kinetochore nor interpolar microtubules, unlike in mitotic mouse fibroblasts or female meiotic oocyte spindles.
