Article Abstract

Intravital multiphoton imaging of the tumor milieu allows for the dissection of intricate and dynamic biological processes . Herein, we present a step-by-step protocol for setting up an experimental cancer imaging model that has been optimized for solid tumors such as breast cancer and melanoma implanted in the flanks of mice. This protocol can be utilized for dissecting tumor-immune cell dynamics or other tumor-specific biological questions. Introduction: The neuronally differentiated key sensory and regulatory cells of the macula densa (MD) were recently identified to control resident progenitor cells and endogenous kidney tissue remodeling and regeneration. Among the many MD-derived secreted angiogenic, growth, and extracellular matrix remodeling factors, cell communication network 1 (CCN1) is a known positive modulator of angiogenesis. Understanding brain function and pathology requires observation of cellular dynamics within intact neural circuits. Background: The perivascular conduct pathway that connects the cerebrospinal fluid spaces with the interstitial fluid in the parenchyma are of importance for solute clearance from the brain. In this pathway, the relatively wide perivascular space (PVS) surrounding the pial arteries provides a low-resistant passage while around the perforating arteries, the solute movement is along the basement membrane (BM), that prevents the free exchange of interstitial fluids and solutes. We hypothesize that this selectivity involves specific components of the vascular BM, which is mainly composed of type IV collagen (Col IV) and laminin networks interconnected by nidogens and heparan sulphate proteoglycans (HSPGs).
