Aggregation of α-synuclein (αS) leads to the hallmark neuropathology of Parkinson's disease (PD) and related synucleinopathies. αS has been described to exist in both cytosolic and membrane-associated forms, the relative abundance of which has remained unsettled. To study αS under the most relevant conditions by a quantitative method, we cultured and matured rodent primary cortical neurons for >17 days and determined αS cytosol:membrane distribution via centrifugation-free sequential extractions based on the weak ionic detergent digitonin. We noticed that at lower temperatures (4 °C or room temperature), αS was largely membrane-associated. At 37 °C, however, αS solubility was markedly increased. In contrast, the extraction of control proteins (GAPDH, cytosolic; calnexin, membrane) was not affected by temperature. When we compared the relative distribution of the synuclein homologs αS and β-synuclein (βS) under various conditions that differed in temperature and digitonin concentration (200-1200 μg/ml), we consistently found αS to be more membrane-associated than βS. Both proteins, however, exhibited temperature-dependent membrane binding. Under the most relevant conditions (37 °C and 800 μg/ml digitonin, i.e., the lowest digitonin concentration that extracted cytosolic GAPDH to near completion), cytosolic distribution was 49.8% ± 9.0% for αS and 63.6% ± 6.6% for βS. PD-linked αS A30P was found to be largely cytosolic, confirming previous studies that had used different methods. Our work highlights the dynamic nature of cellular synuclein behavior and has important implications for protein-biochemical and cell-biological studies of αS proteostasis, such as testing the effects of genetic and pharmacological manipulations.

N-Heterocyclic carbenes (NHCs) hold a unique significance in organometallic catalysis and are powerful organocatalysts for a variety of organic transformations involving crucial intermediates such as Breslow intermediates (BIs), deprotonated BIs (BI-s), ketyl radicals (KRs), and acyl azoliums (AAs). To address the remaining challenges facing reactions catalyzed by NHCs, non-classical stable carbenes, namely 1,2,3-triazolylidenes (MICs), cousins of NHCs, have shown great potential. MICs share similar features with typical NHCs but possess unique characteristics, such as enhanced σ-donor ability and absence of dimerization.

Background And Aims: Data comparing the location of polyp yield in patients with positive stool tests can aid screening test selection. We conducted a cross sectional analysis of New Hampshire Colonoscopy Registry data to compare the location, left versus right side of the colon, of neoplasia detected on colonoscopy following a mt-sDNA+ or FIT+ test as compared to a reference group having colonoscopy without a stool test.

MicroRNAs (miRNAs) are essential regulators of gene expression, yet few comprehensive databases exist for miRNA expression in non-model species, limiting our ability to characterize their roles in gene regulation, development, and disease. Similarly, isomiRs - length and sequence isoforms of canonical miRNAs with potentially altered regulatory targets and functions - have received even less attention in non-model species, including the horse, leaving a critical gap in our understanding of their biological significance. To address these challenges, we developed an open-source, containerized pipeline for identifying and quantifying miRNAs and isomiRs (FARmiR: Framework for Analysis and Refinement of miRNAs), and an associated interactive browser (AIMEE: Animal IsomiR and MiRNA Expression Explorer).

Mental fatigue (MF) is a complex phenomenon with significant implications for human performance, for which there are numerous studies investigating the effects of MF. Nevertheless, there is considerable variability in the approaches used to induce and quantify MF, making it hard to compare findings across studies and draw well-supported conclusions. This review addresses the methodological variability in the induction and quantification methods of MF in movement science in the following ways: on the one hand, by providing an overview of task design strategies to induce MF, emphasizing the importance of tailoring task duration, difficulty, and nature to individual participants and specific research contexts; on the other hand, by providing an overview of current methods used to quantify MF, including behavioural, subjective, and physiological measures, and highlighting the strengths and limitations of each.

Background: Misuse of anabolic-androgenic steroids (AAS), especially through "stacking" multiple substances, poses significant health risks. This study leverages data from the FDA's Adverse Event Reporting System (FAERS) to assess these risks and identify factors predicting severe outcomes.
Methods: We analyzed 286 FAERS reports of intentional AAS misuse.
