Binding of podoplanin to the C-type lectin-like receptor 2 (CLEC-2) promotes platelet activation and soluble CLEC-2 (sCLEC-2) is shed from activated platelets. The role of sCLEC-2 in the plasma is unknown. The expression level and plasma concentration of sCLEC-2 could be affected by variants of the corresponding gene, . Here, we genotyped SNVs in the promoter and coding region of and determined plasma levels of sCLEC-2 in healthy individuals. We genotyped 516 healthy blood donors for 7 SNVs (rs10505743, rs11053538, rs4764178, rs76016091, rs2273986, rs2273987, rs521040) by using PCR methods and calculated haplotypes from the SNV genotypes. For 313 of the donors we measured the sCLEC-2 concentration in EDTA plasma samples by using a commercial ELISA. SNV typing revealed allele frequencies comparable to database information. None of the SNVs showed significant correlation with sCLEC-2 plasma levels. Haplotype analysis indicated 6 haplotypes with frequencies >1% and haplotype h3 was the most frequent (33.8%). Donors homozygous for h3 (n = 37) showed significantly lower sCLEC-2 plasma levels (median 0.95 ng/mL) than donors being h3 negative or heterozygous (n = 276; 1.44 ng/mL; = .0203). We found that the sCLEC-2 plasma concentration is variable in healthy individuals and the genotype contributes to the expression level.

Background: Parasite antigens and plasma lipopolysaccharide (LPS) levels from luminal origin in visceral leishmaniasis (VL) patients are correlated with cellular activation and low CD4+T cell counts.
Objectives: Our aim was to verify whether Leishmania infantum infection damages the intestinal barrier and whether combination antimonial/antibiotic contributes to the reduction of LPS levels and immune activation.
Methods: Golden hamsters were grouped in: G1-uninfected; G2-infected with L.

Epidermal growth factor receptor (EGFR) dimerization plays a pivotal role in cellular signaling, influencing proliferation and disease progression, particularly in cancer. Despite extensive studies, the quantitative relationship between EGFR expression levels and dimerization efficiency remains incompletely understood. In this study, we investigated EGFR dimerization kinetics using ensemble-level biochemical assays and single-molecule tracking (SMT) in living cells.

The legalization of cannabis in several states across the United States has increased the need to better understand its effects on the body, brain, and behavior, particularly in different populations. Previous rodent studies have revealed age and sex differences in response to injected Δ-tetrahydrocannabinol (THC). However, the pharmacokinetic and pharmacodynamic properties of THC administered through more translationally relevant routes of administration are less well known.

Background: Excess aldosterone of > 15ng/dL, in the presence of low renin, is linked to hypertension (HTN) and chronic kidney disease (CKD). This study investigated the association of aldosterone dysregulation at lower plasma aldosterone levels (≥5ng/dL) with the risk of uncontrolled HTN and CKD prevalence.
Methods: Patient plasma aldosterone measurements obtained during 2013-2023 were identified in the TriNetX Dataworks-USA Network of electronic medical records.

Introduction: Diabetes Mellitus is a chronic disease characterised by elevated plasma glucose (PG) levels. HbA1c has been widely utilized for diabetes diagnosis. However, certain conditions restrict its use.
