The mitochondrial GTP (mtGTP)-dependent phosphoenolpyruvate (PEP) cycle couples mitochondrial PEPCK (PCK2) to pyruvate kinase (PK) in the liver and pancreatic islets to regulate glucose homeostasis. Here, small molecule PK activators accelerated the PEP cycle to improve islet function, as well as metabolic homeostasis, in preclinical rodent models of diabetes. In contrast, treatment with a PK activator did not improve insulin secretion in pck2 mice. Unlike other clinical secretagogues, PK activation enhanced insulin secretion but also had higher insulin content and markers of differentiation. In addition to improving insulin secretion, acute PK activation short-circuited gluconeogenesis to reduce endogenous glucose production while accelerating red blood cell glucose turnover. Four-week delivery of a PK activator in vivo remodeled PK phosphorylation, reduced liver fat, and improved hepatic and peripheral insulin sensitivity in HFD-fed rats. These data provide a preclinical rationale for PK activation to accelerate the PEP cycle to improve metabolic homeostasis and insulin sensitivity.

Along their displacement trajectory, displaced adolescent girls and young women (AGYW) face elevated HIV risk early in their sexual life course, often due to forced sexual initiation (FSI), marking the beginning of cycles of violence. However, knowledge gaps exist regarding FSI prevalence rates and the association between FSI, violence experiences, and post-exposure prophylaxis (PEP) cascades (awareness, access and uptake) among displaced AGYW in Uganda. Using peer-driven sampling, we conducted a community-based cross-sectional survey of 201 sexually active displaced AGYW living in informal settlements in Kampala.

Phosphoenolpyruvate carboxykinase (PEPCK) is involved in the conversion of phosphoenolpyruvate (PEP) to oxaloacetate (OAA). In addition to playing a role in gluconeogenesis in various organisms, PEPCK also functions in the C cycle to concentrate CO for photosynthesis in some C plants. Brown algae harbor genes related to the C cycle, including the PEPCK gene, and are proposed to employ a C cycle-like pathway.

The transmembrane glycoprotein known as CD38 (cluster of differentiation 38) is a desirable target for targeted therapeutics as it is essential for immune response and cell signaling, especially in hematological malignancies. The current study aimed to design antibody/nanobody-derived peptides using bioinformatic tools and experimentally investigate their specific binding and impact on the extracellular CD38 expressing cells. Following the synthesis of one of the peptides that indicated somewhat better in silico results, known as CDR3a23 (L-pep), and its circular version as C-pep, flow cytometry was used to assess each one's specific binding to CD38.

Introduction: In 2023, over 210,000 new HIV acquisitions occurred in Kenya, Mozambique, Nigeria, Uganda and Zambia. While uptake of oral pre-exposure prophylaxis (oral PrEP) and coverage of voluntary medical male circumcision increased significantly over the past decade, post-exposure prophylaxis (PEP) has received less attention and remains an underused HIV prevention intervention. In 2024, the World Health Organization (WHO) released new guidance emphasizing the need for timely access to PEP, including through community-based channels and task-sharing to mitigate barriers such as stigma and ensure timely access.
