Article Abstract

New cisplatin analogs in which the diamminedichloro-Pt(II) unit is conjugated to dihydroquinoline- or tetrahydroquinoline frameworks were synthesized and subjected to biological evaluation in order to understand their effects on cellular redox homeostasis and cell viability. They exhibited better selectivity towards cancer cells (A549) compared to mice fibroblast NIH3T3 cells, with cytotoxicity in the same range as that of cisplatin. There was structure-dependent variation in the levels of ROS and were also able to induce cell death, as evidenced by accumulation of cells in sub-G1 phase.

Two new complexes [Pt(L1)Cl]Cl () and [Pt(L2)Cl]Cl (), (L1 = ,-bis(5-methylthiazol-2-yl)pyridine-2,6-dicarboxamide and L2 = ,-di(benzo[]thiazol-2-yl)pyridine-2,6-dicarboxamide), were synthesized and characterized. The rate of nucleophilic substitution reactions of complexes with l-methionine (l-Met), l-cysteine (l-Cys), and guanosine-5'-monophosphate (5'-GMP) decreases in the order of biomolecules reactivity: l-Met > l-Cys >5'-GMP. The complexes exhibited moderate binding affinity for calf thymus DNA and human serum albumin.

An efficient protocol was developed for the microwave-mediated metallation of 5-(4-methoxycarbonylphenyl)-10,15,20-tris(pentafluorophenyl)porphyrin () with bis(benzonitrile)platinum dichloride salt and subsequent 1,3-dipolar cycloaddition of the resulting with an azomethine ylide to give two isomeric metallochlorins: (main isomer) and . The methyl ester group of metalloporphyrin and metallochlorin was successfully hydrolysed in an alkaline medium to yield the corresponding derivatives and in moderate-to-good yields. As a proof of concept of the reactivity of the carboxy group in and , these compounds were conjugated with a hydroxylated derivative of indomethacin, a known potent non-steroidal anti-inflammatory, obtaining the conjugates - and -.

Limited curative therapies can effectively prevent the most malignant form of breast cancer, triple-negative breast cancer (TNBC), from growing and multiplying. Metal-NHC complexes offer a robust foundation for designing an innovative anticancer pharmacophore. Herein, we report the synthesis and characterization of Pd (2), Pt (3), and Au (4) complexes containing pincer (N^C^N) N-heterocyclic carbene ligands derived from 1,3-bis-(1-methyl-1-benzo[]imidazol-2-yl-methyl)-1-imidazol-3-ium hexafluorophosphate (1·HPF).

Compared with Pt analogues that exhibit unique stimulus-induced switching luminescence properties and novel material applications, the properties and reactivity of the MMLCT excited state of Pd complexes in solutions are under-developed. Here, we prepared a series of binuclear cyclometalated Pd complexes with short intramolecular Pd-Pd distances of 2.79-2.

Tumor-Specific On-Site Activation of Cisplatin via Cascade Catalytic-Redox Reactions for Highly Efficient Chemo-Immunotherapy.

Angew Chem Int Ed Engl

The therapeutic efficiency of platinum drugs is always limited by low utilization, side effects, and Pt-resistance. Herein, a double-lock protected Pt nanomedicine named PtNP@Cu has been developed, which performs cascade unlocking of dechlorinated cisplatin (DP) via catalytic-redox reactions, thus achieving tumor-specific "on-site" activation of cisplatin (cDDP) in the nucleus accompanied with substantial induction of ferroptosis of cancer cells. This design avoids the premature release of active Pt species in normal cells or in the cytoplasm of cancer cells before reaching nucleus, thereby ensuring maximum amplification of Pt-DNA crosslinking with tumor-specificity.
