The conventional "one-size-fits-all" approach to treating and managing type 2 diabetes mellitus (T2DM) has proven challenging. T2DM is a complex, heterogeneous disease and frequently coexists with other conditions like hypertension, obesity, and dyslipidemia, further complicating treatment decisions and outcomes. While advances in diabetes medications have been made, traditional treatment algorithms often focus on broad glycemic targets, neglecting the diverse needs of patients with different risk profiles associated with ethnicity, comorbidities, and preferences.

Since Hans Driesch's pioneering work in 1891, it has been known that animal embryos can develop into complete individuals even when divided. However, the developmental processes and molecular mechanisms enabling this self-organization remain poorly understood. In this study, we revisit Driesch's experiments by examining the development of isolated 2-cell stage blastomeres in the sea urchin, Hemicentrotus pulcherrimus.

Objective: This post hoc study of the Progesterone for Traumatic Brain Injury, Experimental Clinical Treatment (ProTECT) III trial investigates whether improving traumatic brain injury (TBI) classification, using serum biomarkers (glial fibrillary acidic protein [GFAP] and ubiquitin carboxyl-terminal esterase L1 [UCH-L1]) and algorithmically assessed total lesion volume, could identify a subset of responders to progesterone treatment, beyond broad measures like the Glasgow Coma Scale (GCS) and Glasgow Outcome Scale-Extended (GOS-E), which may fail to capture subtle changes in TBI recovery.
Design: Brain lesion volumes on CT scans were quantified using Brain Lesion Analysis and Segmentation Tool for CT. Patients were classified into true-positive and true-negative groups based on an optimization scheme to determine a threshold that maximizes agreement between radiological assessment and objectively measured lesion volume.

The classical models of intra-Golgi transport envision a movement of cargoes from cis- to trans-Golgi, followed by their sorting at the trans-Golgi network (TGN). During this vectorial transport, the cargoes are processed by sequentially acting glycosylation enzymes. A number of studies challenged the vectorial transport model and proposed alternative transport routes bypassing either directional transport or the TGN.

Secretory granules are specialized vesicles that package large amounts of cargo, such as hormones, enzymes and mucous. They are typically stored intracellularly until an extracellular signal initiates exocytosis, characterized by fusion between the secretory vesicles and the plasma membrane, resulting in cargo release to the environment. Observations made in the 1970s suggested that some secretory granules project a pseudopodium from their surface where fusion with the cell surface occurs.
