Foot-and-mouth disease virus (FMDV), a critical pathogen in the global livestock industry, has long been a focal point of international disease control strategies. This study developed a nanoparticle-based FMDV vaccine platform. We fused the FMDV immunodominant epitope (VP1-G-H-loop) and T-cell epitope (T) with the nanoparticle scaffold (LS), efficiently producing the T-LS-LOOP nanoparticle vaccine using the prokaryotic expression system (BL21).

Background: Enteroviruses, including Coxsackie B (CVB) viruses, can cause severe diseases such as myocarditis, pancreatitis, and meningitis. Vaccines can prevent these complications, but conserved non-neutralizing epitopes in the viral capsid may limit their effectiveness. The immunodominant PALXAXETG motif, located in the VP1 N-terminus, is a highly conserved region in enteroviruses that elicits non-neutralizing antibody responses.

Immunoassays are complementary diagnostic tools in human cystic echinococcosis (CE) despite sensitivity/specificity limitations, and synthetic peptides have been suggested to potentially overcome disadvantages reported for traditional antigens. Herein, a systematic study comparing the immunodiagnostic performance of AgB1 versus synthetic peptides derived from its sequence was carried out. Thus, a eukaryotic-expressed recombinant AgB1 was assessed, together with a reported synthetic peptide (p176, N-terminal portion of AgB1) and two new peptides within p176 (namely pB1a and pB1b) corresponding to predicted linear B-cell epitopes.

mRNA vaccines emerged as a leading vaccine technology during the COVID-19 pandemic. However, their sustained protective efficacies were limited by relatively short-lived antibody responses and the emergence of SARS-CoV-2 variants, necessitating frequent and variant-updated boosters. We recently developed the ESCRT- and ALIX-binding region (EABR) mRNA vaccine platform, which encodes engineered immunogens that induce budding of enveloped virus-like particles (eVLPs) from the plasma membrane, thereby resulting in presentation of immunogens on cell surfaces and eVLPs.
