Berberine (BR) acts as an AMP-activated protein kinase (AMPK) activator which possesses antioxidant and anti-inflammatory properties. In this study, we have investigated the effects of BR against cerulein-induced chronic pancreatitis (CP) via inhibition of TGF-β/Smad signaling and M2 macrophages polarization in AMPK dependent manner. Cerulein-induced CP mice were treated with BR (3 and 10 mg/kg), intraperitoneally every day for 21 days. Our results indicated that, BR treatment (10 mg/kg) significantly reduced oxidative-nitrosative stress, histological alterations, inflammatory cells infiltration and collagen deposition in pancreatic tissue. BR treatment also prevented cerulein-induced pancreatic stellate cells (PSCs) activation and extracellular matrix (ECM) deposition via downregulation of α-SMA, collagen1a, collagen3a and fibronectin expression. Mechanistically, treatment with BR significantly activated AMPK signaling as compared to cerulein-challenged mice. Further, administration of BR also inhibited TGF-β/Smad signaling and macrophages polarization in cerulein-induced CP in-vivo models and TGF-β1 stimulated RAW 264.7 macrophages in-vitro. Together, our results strongly suggest that BR treatment protected against cerulein-induced CP and associated fibrosis progression by inhibiting TGF-β1/Smad signaling and M2 macrophages polarization in an AMPK dependent manner.

Macrophage Migration Inhibitory Factor (MIF) is a pleiotropic cytokine that acts as a central regulator of inflammation and immune responses across diverse organ systems. Functioning upstream in immune activation cascades, MIF influences macrophage polarization, T and B cell differentiation, and cytokine expression through CD74, CXCR2/4/7, and downstream signaling via NF-κB, ERK1/2, and PI3K/AKT pathways. This review provides a comprehensive analysis of MIF's mechanistic functions under both physiological and pathological conditions, highlighting its dual role as a protective mediator during acute stress and as a pro-inflammatory amplifier in chronic disease.

Objectives: In this study, we explored the mechanism by which DDIT4 influences the polarization phenotypic transformation of macrophages and inflammation through the regulation of mTOR signaling pathway, providing a new mechanism and target for the treatment of diabetic nephropathy.
Methods: The degree of inflammation and injury in renal tissues of diabetic kidney disease (DKD) animal model was evaluated using biochemical assays, renal pathology examinations, and Western blot tests. Podocytes and macrophages were isolated from renal tissues to observe the extent of podocyte injury and the quantity and polarization phenotype of macrophage infiltration.

Background: Atherosclerosis (AS) is a leading risk factor for cardiovascular diseases globally, characterised by the accumulation of lipids and cholesterol in arterial walls, causing vascular narrowing and sclerosis along with chronic inflammation; this leads to increased risk of heart disease and stroke, significantly impacting patients' health. Danxia Tiaoban Decoction (DXTB), a traditional Chinese medicine (TCM) formula, has demonstrated positive clinical effects in treating AS; however, its mechanisms of action remain unclear.
Objective: To explore the potential mechanisms of action of DXTB in treating AS through multi-omics integration and experimental validation.

Seven previously undescribed glycosides, including four iridoid glycosides (1, 2, 4, and 5), three phenolic glucosides (18, 21, and 22), together with nineteen known compounds, were isolated from the ethanol extract of Gentiana rhodantha Franch. Their structures were determined by analysis of 1D and 2D NMR, HRESIMS, X-ray diffraction, NMR calculation, and electronic circular dichroism (ECD) calculation. Among them, compound 21 was a novel highly rearranged endo-phenolic analogue with a unique skeleton.
