Purpose: Precision medicine has been most successful in targeting single mutations, but personalized medicine using broader genomic tumor profiles for individual patients is less well developed. We evaluate a genomics-informed computational biology model (CBM) to predict outcomes from standard treatments and to suggest novel therapy recommendations in glioblastoma (GBM).
Methods And Materials: In this retrospective study, 98 patients with newly diagnosed GBM undergoing surgery followed by radiation therapy and temozolomide at a single institution with available genomic data were identified. Incorporating mutational and copy number aberration data, a CBM was used to simulate the response of GBM tumor cells and generate efficacy predictions for radiation therapy (RTeff) and temozolomide (TMZeff). RTeff and TMZeff were evaluated for association with overall survival and progression-free survival in a Cox regression model. To demonstrate a CBM-based individualized therapy strategy, treatment recommendations were generated for each patient by testing a panel of 45 central nervous system-penetrant US Food and Drug Administration-approved agents.
Results: High RTeff scores were associated with longer survival on univariable analysis (P < .001), which persisted after controlling for age, extent of resection, performance status, MGMT, and IDH status (P = .017). High RTeff patients had a longer overall survival compared with low RTeff patients (median, 27.7 vs 14.6 months). High TMZeff was also associated with longer survival on univariable analysis (P = .007) but did not hold on multivariable analysis, suggesting an interplay with MGMT status. Among predictions of the 3 most efficacious combination therapies for each patient, only 2.4% (7 of 294) of 2-drug recommendations produced by the CBM included TMZ.
Conclusions: CBM-based predictions of RT and TMZ effectiveness were associated with survival in patients with newly diagnosed GBM treated with those therapies, suggesting a possible predictive utility. Furthermore, the model was able to suggest novel individualized monotherapies and combinations. Prospective evaluation of such a personalized treatment strategy in clinical trials is needed.

Several approaches for synthesizing [1-C]2-oxoglutaric acid were attempted, and the synthesis was successfully achieved in 4 steps from trimethylsilyl C-cyanide. The C-breath tests on rats were conducted by orally administering the newly synthesized [1-C]2-oxoglutaric acid, the previously prepared [1'-C]citric acid, and [1-C]acetic acid as a control drug, and the results were compared. The results indicate that [1-C]2-oxoglutaric acid and [1'-C]citric acid may serve as potential substrates for assessing the TCA cycle flux.

Background: Previous studies have operationalized the NCCN list of high-risk medications in older adults into a measurable tool known as the Geriatric Oncology Potentially Inappropriate Medications (GO-PIMs) scale. The current study aims to evaluate the ability of GO-PIMs to identify high-risk medications and their impact on patients with both solid and liquid tumors managed in a large national health care system.
Methods: We performed a retrospective cohort study using data from the national Veterans Affairs (VA) Cancer Registry and electronic health records, including all veterans newly diagnosed with a solid or liquid malignancy from 2000 to 2022.

This study aimed to histomorphometrically evaluate the effect of guided bone regeneration (GBR) and two implant surfaces on the thickness and height of newly formed bone in dehiscence defects around titanium implants. Three premolars and the first bilateral molar were extracted from ten adult mongrel dogs, and 40 buccal bone dehiscences measuring 5 mm in height and 4 mm in width were created using a University of North Carolina (UNC) periodontal probe to confirm the dimensions. Forty implants were randomly assigned to one of four groups: oxidized implant surfaces (OIS, n = 10), turned/machined implant surfaces (TIS, n = 10), OIS + GBR (n = 10), and TIS + GBR (n = 10).
