Matrix Gla protein (MGP), an extracellular matrix protein, is mainly associated with the inhibition of calcification in skeleton, coronary artery, and kidney, and more recently it has also been implicated in cancer. However, the biological function of MGP inside cancer cells and its role in colon cancer (CC) remain largely unknown. MGP expression and its association with clinicopathologic characteristics in CC were analyzed by immunohistochemistry and verified by Gene Expression Omnibus (GEO) and The Cancer Genome Atlas (TCGA) datasets. The effects of MGP on CC cell proliferation were evaluated via knockdown and overexpression experiments . Mechanisms of MGP in CC were explored by western blots, quantitative real-time PCR, Fluo-3 AM staining, Rhod-2 AM staining, immunofluorescence, and other techniques. Our study confirmed that MGP was upregulated in different stages of CC and associated with a worse prognosis. MGP could enrich intracellular free Ca concentration and promote nuclear factor κB (NF-κB)/p65 phosphorylation, activating the expression of c-MYC, ICAM-1, and VEGFA. Furthermore, the reduction of intracellular free Ca concentration and the subsequent growth inhibition effect on CC cells induced by small interfering RNA targeting MGP (siMGP) could be rescued by a higher calcium concentration environment. Therefore, MGP promotes the growth and proliferation of CC cells by enriching intracellular calcium concentration and activating the NF-κB pathway, and it could serve as a potential prognostic biomarker in CC patients.

Background: Patients with primary membranous nephropathy may progress to advanced chronic kidney disease despite immunosuppressive therapy (IST). Prediction of treatment response based on early and combined assessment of several standard clinical markers could improve risk stratification for progression, allowing timely individualization of treatment, which can optimize clinical outcomes and safety.
Methods: In this exploratory analysis of the STARMEN trial, we evaluated if combined baseline data, and IST-induced early changes in standard clinical markers predicted clinical remission at 2 years.

To assess the association between dietary consumption patterns of antioxidant and pro-oxidant nutrients with prostate cancer (PC) and its histological differentiation, we analyzed data from 394 histologically confirmed incident cases of PC and 793 age-matched population controls (± 5 years), residents of Mexico City. Cases were classified by Gleason score into well-, moderately-, and poorly differentiated categories. Dietary nutrient intake over the three years preceding diagnosis for cases and before the interview for controls was estimated using a semi-quantitative food frequency questionnaire.

Brivaracetam (BRV) and levetiracetam (LEV) are antiseizure medications (ASMs); UCB-J is a PET tracer targeting synaptic vesicle protein 2A (SV2A); UCB7361 is closely related to padsevonil, an experimental anticonvulsant; while UCB1244283 acts as an allosteric modulator for BRV and LEV binding but not for these other ligands. The SV2A-BRV-UCB1244283 structure reveals how UCB1244283 allosterically enhances BRV binding by occupying an allosteric site near the primary binding site, preventing BRV dissociation. This allosteric site, formed by hydrophobic and uncharged residues, is an uncharacterized small-molecule binding site in SV2A.

Livestock and poultry farming effluents (LPFE) have become a significant source of pollution in the agricultural water environment, with nitrogen (N) and phosphorus (P) being the major pollutants. The use of inexpensive Mg2+ -modified materials as crystallization carriers can enhance the struvite crystallization and precipitation reaction, enabling efficient recovery of N and P nutrients from LPFE. In this study, Mg2+ -modified corncob biochar (CCBC-Mg) and Mg2+ -modified natural zeolite (NZ-Mg) were assessed for their simultaneous recovery of N and P from LPFE.
