(1) Background: Therapeutic blocking of the interaction between programmed death-1 (PD-1) with its ligand PD-L1, an immune checkpoint, is a promising approach to restore the antitumor immune response. Improved clinical outcomes have been shown in different human cancers, including non-small cell lung cancer (NSCLC). Unfortunately, still a high number of NSCLC patients are treated with immunotherapy without obtaining any clinical benefit, due to the limitations of PD-L1 protein expression as the currently sole predictive biomarker for clinical use; (2) Methods: In this study, we applied mass spectrometry imaging (MSI) to discover new protein biomarkers, and to assess the possible correlation between candidate biomarkers and a positive immunotherapy response by matrix-assisted laser desorption/ionization (MALDI) MSI in 25 formalin-fixed paraffin-embedded (FFPE) pretreatment tumor biopsies (Biobank@UZA); (3) Results: Using MALDI MSI, we revealed that the addition of neutrophil defensin 1, 2 and 3 as pretreatment biomarkers may more accurately predict the outcome of immunotherapy treatment in NSCLC. These results were verified and confirmed with immunohistochemical analyses. In addition, we provide in-vitro evidence of the immune stimulatory effect of neutrophil defensins towards cancer cells; and (4) Conclusions: With proteomic approaches, we have discovered neutrophil defensins as additional prospective biomarkers for an anti-PD-(L)1 immunotherapy response. Thereby, we also demonstrated that the neutrophil defensins contribute in the activation of the immune response towards cancer cells, which could provide a new lead towards an anticancer therapy.

β-defensins are multifunctional peptides of the host immune system involved in responses to infectious diseases. We investigated the potential of five recombinant proteins based on bovine β-defensins (bovine neutrophil β-defensins (BNBD) 1, 2, 3, and 4, and the tracheal antimicrobial peptide (TAP)) in functions relevant to sepsis such as antimicrobial activity, lipopolysaccharide (LPS) binding and neutralisation, and the stimulation of cytokine response in epithelial cells. These β-defensins were produced in Lactococcus lactis as fusion proteins.

Aims: The study aims to assess the gut microbiota function and the status of bidirectional relationship of gut with other body organs in ulcerative colitis (UC).
Methods And Results: The proteomic analysis of fecal samples revealed the presence of 2072 microbial proteins and 243 human proteins. Microbial proteins for pentose phosphate pathway (PPP) increased significantly in UC samples; whereas, decrease in proteins related to energy metabolism, transmembrane transporters and membrane proteins, and protein folding was noted.

Neutrophils play a critical role in the first-line of defense against circulating pathogens and contain a wide array of granules that store antimicrobial proteins, with neutrophil serine proteases (NSPs) and defensins serving as crucial components. NSPs such as neutrophil elastase (NE), proteinase 3 (PR3), cathepsin G (CatG) and neutrophil serine protease 4 (NSP4) exhibit distinct substrate specificities that underpin their critical roles in immune defense and inflammation [1]. After neutrophils are activated, they form and release neutrophil extracellular traps (NETs) consisting of decondensed chromatin and intracellular proteins through a process called NETosis, which leads to neutrophil death.
