DNA extracted from formalin-fixed, paraffin-embedded tissue sections is often inadequate for sequencing, due to poor yield or degradation. We optimized the proteinase K digest by testing increased volume of enzyme and increased digest length from the manufacturer's protocol using 54 biospecimens, performing the digest in centrifuge tubes. Doubling the quantity of proteinase K resulted in a median increase in yield of 96%. Applying the optimized proteinase K protocol to sections deparaffinized on microscope slides generated a further increase in yield of 41%, but only at >50,000 epithelial tumor cells/section. DNA yield now correlated with (χ = 0.84) and could be predicted from the epithelial tumor cell number. DNA integrity was assayed using end point multiplex PCR (amplicons of 100-400 bp visualized on a gel), quantitative PCR (qPCR; Illumina FFPE QC Assay), and nanoelectrophoresis (DNA Integrity Numbers [DINs]). Generally, increases in yield were accompanied by increases in integrity, but sometimes qPCR and DIN results were conflicting. Amplicons of 400 bp were almost universally obtained. The process of optimization enabled us to reduce the percentage of samples that failed published quality control thresholds for determining amenability to whole genome sequencing from 33% to 7%.

Dipeptidyl-peptidase (DPP)-IV inhibition by penultimate N-terminus Pro-containing peptides is a promising strategy for Type 2 diabetes (T2D) management, as it prevents the degradation of incretin hormones (DPP-IV substrates) like glucagon-like peptide-1 (GLP-1), thereby prolonging their half-life. However, the stability and bio-accessibility of these peptides are crucial to their efficacy in orally administered therapeutics. We previously identified LPCL and TPFLPDE peptides from tilapia viscera by-products hydrolysates, which exhibited significant DPP-IV inhibition in vitro and in situ while effectively preserving active GLP-1 levels after 2 h treatment in STC-1 cells under basal glucose conditions.

This study systematically compared the effects of microwave (MW) and conventional thermal (CT) processing on microbial inactivation, enzyme activity, protein retention, and nutritional characteristics of bovine milk under pasteurization (70 °C for 20 s) and ultra-high temperature (UHT) sterilization conditions. Both technologies achieved effective microbial reduction (>4 log CFU/mL) for Escherichia coli, Staphylococcus aureus, and Salmonella spp., complying with EU and FDA safety standards.

Background: The programmed cell death protein 1 (PDCD1 or PD-1) is a key regulatory immune checkpoint and a major target for therapeutic intervention. In oncology, antibodies blocking the PD-1 pathway are used to activate immune cells to promote anti tumour immunity while in immune-mediated inflammatory diseases, PD-1 agonist molecules have the potential to achieve immune suppression. NK cells are a specialised population of innate lymphocytes able to recognize a large range of distressed cells including damaged tissues in autoimmune and inflammatory conditions.
