Interactions of Oral Molecular Excipients with Breast Cancer Resistance Protein, BCRP.

Article Abstract

Mechanistic-understanding-based selection of excipients may improve formulation development strategies for generic drug products and potentially accelerate their approval. Our study aimed at investigating the effects of molecular excipients present in orally administered FDA-approved drug products on the intestinal efflux transporter, BCRP (), which plays a critical role in drug absorption with potential implications on drug safety and efficacy. We determined the interactions of 136 oral molecular excipients with BCRP in isolated membrane vesicles and identified 26 excipients as BCRP inhibitors with IC values less than 5 μM using H-cholecystokinin octapeptide (H-CCK8). These BCRP inhibitors belonged to three functional categories of excipients: dyes, surfactants, and flavoring agents. Compared with noninhibitors, BCRP inhibitors had significantly higher molecular weights and SLogP values. The inhibitory effects of excipients identified in membrane vesicles were also evaluated in BCRP-overexpressing HEK293 cells at similar concentrations. Only 1 of the 26 inhibitors of BCRP identified in vesicles inhibited BCRP-mediated H-oxypurinol uptake by more than 50%, consistent with the notion that BCRP inhibition depends on transmembrane or intracellular availability of the inhibitors. Collectively, the results of this study provide new information on excipient selection during the development of drug products with active pharmaceutical ingredients that are BCRP substrates.

The development of analytical techniques applicable to powdered pharmaceutical co-crystals, including those containing excipients, represents a comprehensive strategy for quality control in both drug development and regulatory settings. This study investigates the structural characterization of indomethacin-nicotinamide co-crystals using a combination of microcrystal electron diffraction (microED), solid-state NMR (SSNMR), Raman spectroscopy, and powder X-ray diffraction (PXRD). MicroED analysis revealed the crystal structure of the co-crystal, while SSNMR measurements provided insights into the molecular interactions within the structure.

Gout, which affects 3-6 % of Western populations, has well-established therapies but still lacks agents that directly target monosodium urate (MSU) deposits. This study investigates a novel strategy employing cyclodextrins (CDs) and hyperbranched cyclodextrin-based polymers (HBCD-Pol) to both mobilize and prevent MSU formation. Among the CDs tested, HPβ-CD exhibited the strongest uric acid (UA) complexation at 25 °C, while HBCD-Pol showed superior performance by chelating Na ions.

Small molecules can affect a protein's fold and stability. Physical chemistry conceptualizes this effect using Tanford's transfer model, which describes the change in free energy of a protein upon transfer from water to a solution containing some concentration of a small molecule. Using additive transfer free energies of individual amino acids, one can apply the transfer model to quantitatively predict how small molecules affect protein folding equilibria.
