Background: Resistance to epidermal growth factor receptor tyrosine kinase inhibitors (EGFR-TKIs), such as gefitinib, is a limited factor in the treatment of non-small-cell lung cancer (NSCLC) patients. Therefore, ongoing studies are trying to identify EGFR-TKIs-resistant mechanisms and to discover novel therapeutic strategies and targets for NSCLC treatment.
Methods: In the present study, the possibility of overcoming intrinsic gefitinib-resistance was examined by regulating the expression of AXL. A natural product-derived antitumor agent, yuanhuadine (YD) was employed to modulate the expression of AXL in the cells.
Results: Treatment with YD effectively downregulated AXL expression in AXL-overexpressed gefitinib-resistant H1299 cells. The combination of gefitinib and YD exhibited a synergistic grwoth-inhibitory activity in H1299 cells by downregulation of AXL expression.
Conclusions: Based on these findings, AXL was found to be a promising therapeutic target to overcome the intrinsic resistance to gefitinib in NSCLC. Furthermore, YD is able to effectively regulate the expression of AXL and thus it may be applicable as a potential lead compound for the treatment of gefitinib-resistant NSCLC.

Collecting duct carcinoma (CDC) is a rare subtype of renal cell carcinoma with a poor prognosis. Moreover, despite various chemotherapeutic strategies and administration of several tyrosine kinase inhibitors for metastatic CDC, the outcomes remain unfavorable, with no established treatment. Herein, we report the cases of two patients with CDC who exhibited a good response to nivolumab and cabozantinib combination therapy.

Ethnopharmacological Relevance: White matter injury (WMI) following ischemic stroke represents a critical pathological determinant of persistent neurological impairment, with current therapeutic options remaining limited. Buyang Huanwu Decoction (BYHWD), a time-honored formulation historically deployed in traditional Chinese medicine to address post-stroke sequelae, exhibits documented neuroprotective efficacy; nevertheless, its mechanistic actions governing post-ischemic white matter restoration and remyelination are yet to be fully deciphered.
Aim Of The Study: This study aimed to elucidate whether BYHWD facilitates post-ischemic white matter restoration via TREM2-dependent mechanisms.

Different risk factors are involved in the initiation and progression of melanoma. In particular, genetic and epigenetic pathways are involved in all stages of melanoma and are exploited in therapeutic approaches. This study investigated the role of circular RNA circ_0001591 in melanoma cell migration.

Introduction: The inflammatory phenotype of acute kidney injury (AKI), characterized by interstitial infiltration of immune cells, arises due to nephrotoxic agents. However, it does not pose the same risk of occurrence and progression for everyone, suggesting that the amplification or attenuation of disease depends on the unique immunological status of each kidney. Here, our study investigated the regulatory role of kidney-resident macrophages (KRMs) in the induction and progression of toxin-induced AKI.

Preeclampsia (PE) is a leading cause of maternal and fetal morbidity that affects 2-8% of pregnancies worldwide, driven by placental dysfunction and systemic inflammation. Growth arrest-specific protein 6 (Gas6) and its receptor AXL play pivotal roles in PE pathogenesis, promoting trophoblast impairment and vascular dysregulation. This study investigated the transcriptomic reversal effects of AXL Receptor Tyrosine Kinase (AXL) inhibition in a Gas6-induced rat model of PE using RNA sequencing (RNA-seq).
